Summary: Bone and bone marrow are common metastatic sites of cancer, often associated with poor disease prognosis. Current therapeutic options for bone metastases are limited and mainly palliative. Further research into the molecular pathways of bone metastasis is needed to develop more successful treatments for improved patient outcomes.

Summary: This review evaluated the effectiveness of electrochemotherapy in the treatment of bone metastases. The findings suggest that electrochemotherapy can be considered as a minimally invasive and tissue-sparing treatment for patients who have no other alternatives for tumor control and improvement in quality of life.

Summary: Bone is a common site for metastasis from various primary tumors, with breast cancer and prostate cancer being the most common. The median survival time for patients with bone metastases is short, emphasizing the need for new targets for diagnosis and treatment. Through the analysis of gene expression in bone metastasis using two data sets, PECAM1 was identified as a potential biomarker. Further experiments showed that PECAM1 may have a role in osteoclast function, and its downregulation promoted osteoclast differentiation and enhanced tumor cell proliferation and migration. These findings suggest that PECAM1 could be a promising target for the diagnosis and treatment of bone metastases.

Summary: Bone metastases are a lethal condition commonly seen in solid tumors such as prostate, breast, lung, and renal cell carcinomas. They lead to skeletal-related events (SREs) including pain, fractures, and spinal cord compression. The unique metastatic niche in bone involves a complex interaction between cancer cells, non-cancerous cells, and signaling pathways, resulting in bone remodeling, immune suppression, and therapeutic resistance. This review provides an overview of the approaches used to study bone metastases, potential therapeutic targets, and current clinical investigations of new drugs. It also discusses the challenges, preclinical models, and signaling pathways associated with bone metastases, as well as opportunities to advance research in this field.

Summary: Bone metastasis is a common phenomenon in lung cancer, prostate cancer, and other malignant tumors. In this study, biomineralized metal-organic framework (MOF) nanoparticles carrying protein toxins and combined with RANKL antibody were constructed to effectively treat bone metastases and reduce skeletal-related events (SREs).

Summary: Thyroid cancer is a common endocrine cancer, and bone metastasis significantly affects survival rates. There is no definitive cure, and current treatments are palliative. This study assessed the incidence and influence of bone metastasis on thyroid cancer patients, identifying risk factors and outcomes that can guide clinical decisions and research.

Summary: This study explores the use of acridine orange radiodynamic therapy (AO-RDT) as a targeted anticancer treatment for bone metastasis. The results demonstrate that AO-RDT is selectively toxic to carcinoma cells, effectively inhibits tumor expansion and tumor-induced bone resorption, without affecting osteoclasts.
