Summary: This study evaluates the value of tissue inhibitors of MMPs-2 (TIMP-2) as an indicator of 5-Fluorouracil (5-Fu) resistance in colorectal cancer. The results show that TIMP-2 expression is elevated in drug-resistant colorectal cancer cell lines and is closely related to 5-Fu resistance. Monitoring serum TIMP-2 levels can help clinicians identify 5-Fu resistance in colorectal cancer patients earlier during chemotherapy.

Summary: The study found that PGP can alleviate collagenase-induced OA in rats by reducing blood ALP levels, inhibiting cartilage erosion, increasing COL-2 gene expression, and decreasing MMP-3 and COX-2 gene expression. PGP's high phenolic and flavonoid content contribute to its strong antioxidant activity, making it a potential preventive treatment for OA that warrants further validation in clinical studies.

Summary: This study utilized a newly developed microfluidic swirl mixer to fabricate temperature-sensitive liposomes (TSL) for the treatment of breast cancer through near-infrared stimulation. The results demonstrated that AP-3/ICG@TSL significantly inhibited the growth of breast cancer both in vitro and in vivo without causing damage to other organs. These findings suggest that the AP-3/ICG@TSL manufactured using the microfluidic swirl mixer is a promising drug delivery system for breast cancer therapy.

Summary: Ectodomain shedding, a post-translational modification, is crucial for cell communication, adhesion, and migration. Metalloproteases, especially ADAMs and MT-MMPs, are the main proteases involved in ectodomain shedding. TIMP-3 acts as a master regulator by inhibiting various families of metalloproteases, including ADAMs and MT-MMPs. This review discusses the functions of TIMP-3 in health and disease, with a focus on its role in controlling ectodomain shedding, and introduces mass spectrometry-based approaches for identifying new functions of sheddases and TIMP-3.

Summary: Osteoarthritis is a chronic degenerative joint disease with no current treatment. Tissue inhibitor of metalloproteinases 3 (TIMP-3) is a potential target for osteoarthritis therapy due to its ability to inhibit cartilage degradation. Suramin and its analogues have been found to block the degradation of TIMP-3, making them potential therapeutic agents for osteoarthritis.

Summary: Apoptosis deficiency is a crucial feature in neoplastic diseases, with the Bcl-2 family of proteins playing a decisive role in regulating apoptosis. Studies have shown that hyperactivation of Bcl-2 anti-apoptotic effects is associated with cancer occurrence and resistance to therapy, making targeting Bcl-2 inactivation a promising route for cancer treatment.

Summary: This study introduces a new prodrug-based self-assembled nanoparticle for efficient cancer treatment. The nanoparticles can be quickly enriched in tumor cells and release the drug, showing satisfactory tumor killing effects with weak toxicity and side effects. These unique features could further advance the research progress of prodrug-based self-assembled strategies for chemotherapy in cancer.

Summary: This study conducted a meta-analysis of multiple case-control studies and found that the polymorphism at the rs1871054 site of the ADAM12 gene is associated with genetic susceptibility to osteoarthritis, while the polymorphisms at the rs3740199, rs1044122, and rs1278279 sites are not significantly related to osteoarthritis.

Summary: The repairing of damaged tissues in the gastric tract starts with extracellular matrix remodeling by matrix metalloproteinases (MMPs). The drug omeprazole, used to cure gastric ulcers, has been shown to restore MMP-2 activity and reduce apoptosis, preventing stress-induced gastric ulcers by modulating NF-kappa B and PPAR-gamma activity.

Summary: This study discovered that HIFs induce the expression of ADAM12, leading to EGFR signaling activation and subsequently promoting breast cancer cell migration, invasion, and metastasis. Inhibition of ADAM12 expression or activity can effectively reduce tumor cell migration, invasion, and lung metastasis.

Summary: Cancer stem cells (CSCs) play a crucial role in tumor progression, metastasis, and recurrence. The epigenetic modification machinery is essential for the viability and aggressive evolution of CSCs. Targeting these mechanisms with specific drugs offers a promising approach to block CSCs and reduce drug resistance, quiescence, and tumor heterogeneity.
