Catecholic motifs are present in numerous natural products and synthetic compounds used in various sectors of the chemical industries such as food, cosmetic, pharmaceutical and polymer industries. The antioxidant activity usually conferred to compounds bearing catecholic motifs is the property on which their application is often based. Although several (bio) chemical methods are available to chemists to produce catechols, the oxygenation of phenols constitutes one of the most practical approaches as long as the ortho-selectivity of the process can be controlled. In this context, oxygenating hypervalent iodine(V) reagents offer a convenient metal-free solution. Among these reagents, 2-iodoxybenzoic acid (IBX) or its stabilized (non explosive) SIBX and polymer-supported versions have found the most useful and successful applications in the conversion of phenols into ortho-quinones, followed by reduction into catechols. Examples of oxygenative demethylation of 2-methoxyphenols and ortho-hydroxylation of phenolic compounds for the synthesis of biologically and industrially-relevant catechols are highlighted in this review article, together with some mechanistic discussions on these transformations.

Polyphenols Extracts from Oil Production Waste Products (OPWPs) Reduce Cell Viability and Exert Anti-Inflammatory Activity via PPAR gamma Induction in Colorectal Cancer Cells

Manuela Leo, Livio Muccillo, Laura Dugo, Roberta Bernini, Luca Santi, Lina Sabatino

Summary: Olive oil production generates waste products rich in water-soluble polyphenols, which can pose environmental problems. However, these waste products can be exploited for their bioactive properties. This study chemically characterized the polyphenolic extracts from the waste products and investigated their biological activities in normal and colorectal cancer cells. The extracts showed anti-inflammatory effects and can potentially ameliorate inflammation associated with diseases and tumors.

Summary: This in vitro study evaluated for the first time the antioxidant and anti-inflammatory effect of an Oleuropein-enriched extract on bovine mammary epithelial cells. The extract showed no cytotoxic effect on the cells and reduced ROS production induced by hydrogen peroxide. It also downregulated the expression of pro-inflammatory cytokines. These findings confirm the antioxidant and anti-inflammatory properties of the extract.

Summary: Inflammaging refers to chronic low-grade inflammation caused by immune and non-immune cells stimulated by endogenous/self, misplaced or altered molecules, belonging to physiological aging. This inflammatory status is characterized by increased inflammation and decreased autophagy, and predisposes to age-related diseases. Phenolic compounds, particularly hydroxytyrosol (HTyr) found in extra-virgin olive oil, have antioxidant and anti-inflammatory properties that may counteract inflammaging and contribute to healthy aging.
