CB1R, CB2R and TRPV1 expression and modulation in in vivo, animal glaucoma models: A systematic review

Summary: This study explores the expression and effects of cannabinoid receptor 1 (CB1R), cannabinoid receptor 2 (CB2R), and transient receptor potential vanilloid 1 (TRPV1) in ocular tissues. Preclinical studies show that these receptors have a widespread distribution in the trabecular meshwork, ciliary body, and retina, and their modulation can lead to ocular hypotensive and neuroprotective effects. However, the exact mechanisms responsible for these effects are still unclear. Further research is needed to determine the potential therapeutic target of the endocannabinoid system in glaucoma.

Requirements for Translation in Clinical Trials of Aromatherapy: The Case of the Essential Oil of Bergamot (BEO) for Management of Agitation in Severe Dementia

Summary: This article discusses the application of aromatherapy in the treatment of dementia, particularly the clinical translation process of bergamot essential oil (BEO). Using a nanotechnology-based delivery system loaded with BEO, it is possible to effectively alleviate pain and agitation in dementia patients. The ongoing BRAINAID trial will evaluate the effectiveness and safety of NanoBEO in treating severe dementia patients.

Summary: Migraine, the leading cause of disability in individuals under 50, has seen a paradigm shift in pharmacotherapy with the emergence of CGRP receptor antagonists known as gepants. This review traces the history of CGRP in migraine and highlights the potential for new generation gepants to modulate nociceptive facilitation pathways. Real-world post marketing data is necessary for a comprehensive understanding of gepants in long-term use.

Summary: This study found that severe cerebral ischemia caused by transient MCAo in adult male mice increased inflammatory neutrophils and monocytes in the blood, as well as miRNA-329-5p levels in the plasma. These inflammatory responses could be prevented by ischemic PC induced by 15 min MCAo before the severe insult.

Efficacy of Essential Oils in Relieving Cancer Pain: A Systematic Review and Meta-Analysis

Summary: More than 80% of cancer patients experience cancer-related pain, which is difficult to manage, especially in the advanced stage. Recent evidence suggests that aromatherapy using essential oils can effectively reduce the intensity of cancer pain. This systematic review and meta-analysis evaluated the efficacy of aromatherapy in clinical studies on cancer pain and found significant benefits. However, more standardized and well-designed clinical trials are needed to establish a stronger evidence base for the use of essential oils in integrative oncology. Safety of Onabotulinumtoxin A in Chronic Migraine: A Systematic Review and Meta-Analysis of Randomized Clinical Trials

Summary: Around 14% of the global population in high-income countries suffer from migraine. A systematic review and meta-analysis was conducted to evaluate the safety of onabotulinumtoxin A treatment for chronic migraine. The findings suggest that while onabotulinumtoxin A produces more treatment-related adverse events (TRAEs) compared to placebo, it is less than oral topiramate, indicating the safety of onabotulinumtoxin A. However, there is a need for further clinical trials to assess its safety with newer treatment options.

Neuroprotective Effect of a Nutritional Supplement Containing Spearmint Extract, Forskolin, Homotaurine and Group B Vitamins in a Mouse Model of Transient Ocular Hypertension

Summary: In a mouse model of acute glaucoma, oral administration of a nutritional supplement containing adenylate cyclase, homotaurine, spearmint extract and vitamins of the B group improved retinal ganglion cell (RGC) survival. These effects were independent from the hypotensive effects of the supplement and were associated with decreased calpain activity, reduced caspase-3 and PARP-1 activation, and prevention of GFAP upregulation.

Scopolamine-Induced Memory Impairment in Mice: Effects of PEA-OXA on Memory Retrieval and Hippocampal LTP

Summary: This study examines the effects of PEA-OXA on scopolamine-induced deficits in working memory, discriminative memory, anxiety, and motor activity. It is found that PEA-OXA can prevent or restore scopolamine-induced cognitive deficits, specifically in discriminative and spatial memory. However, it does not affect the altered motor activity or anxiety-like behavior induced by scopolamine. The study confirms the neuroprotective and pro-cognitive effects of PEA-OXA.
