Summary: Cancer therapy has advanced greatly in the past decade, providing new strategies to inhibit cancer cell growth using immune modulation and gene therapy. Recent advances in mRNA delivery have shown the potential of mRNA-based vaccines and immune-modulators for cancer therapeutics. In our study, we demonstrated that local administration of LNPs entrapping a modified mRNA encoding a bacterial toxin induced significant anti-tumor effects and improved overall survival of treated mice. We propose mmRNA-loaded LNPs as a new class of anti-tumoral, toxin-based therapy.

Summary: RNA interference (RNAi) is a treatment approach used to decrease excessive gene expression in diseases, including cancer. However, challenges such as poor cellular uptake, off-target effects, and susceptibility to destruction restrict the therapeutic potential of small interfering RNAs (siRNAs). This research focuses on sophisticated nanocarriers that aid in the transport of siRNA payloads, their combination with anticancer medications, and their use in cancer treatment. It examines molecular manifestations, transport difficulties, delivery method design and development, and the advantages and disadvantages of various nanocarriers. Challenges such as siRNA trapping in endosomes and techniques for siRNA release are also discussed. The study also provides an evaluation of the current state of siRNA treatments in clinical trials for efficient cancer therapy.
