Summary: The ability to produce IL-2 identifies constituents of the expanded CD8 T cell effector pool with stem-like features, which exhibit superior protective powers. The production of IL-2 by cells attenuates the ability to receive IL-2 signals, limiting terminal effector formation and conferring superior protection. Non-producing effector cells, on the other hand, gain effector traits at the expense of memory formation. Despite distinct properties during the effector phase, IL-2-producing and non-producing CD8 T cells converge transcriptionally as memory matures, forming populations with equal recall abilities.

Summary: This study demonstrates the importance of locally produced IL-23 in promoting in situ proliferation of cutaneous TRM17 cells, supporting their longevity and function. Anti-IL-23 therapy effectively reduces the number of TRM17 cells in the skin, which has significant implications for the treatment of psoriasis.

Batf stabilizes Th17 cell development via impaired Stat5 recruitment of Ets1-Runx1 complexes

Summary: This study reveals that the activator protein-1 (AP-1) factor Batf ensures Th17 cell identity by restraining IL-2 expression and IL-2-induced Stat5 activation, thereby inhibiting the transcription of Th1 and Treg cell-specific genes. In addition to its role in regulating Th17-specific loci, Batf indirectly inhibits the assembly of a Stat5-Ets1-Runx1 complex, which enhances the transcription of Th1 and Treg cell-specific genes. These findings highlight the importance of Stat5-Ets1-Runx1 interactions in determining T cell fate and underscore the crucial role of Batf in both inducing and maintaining the Th17 program.
