Summary: There has been a significant increase in scientific research on chronic traumatic encephalopathy (CTE) in the past 17 years. CTE has been diagnosed in hundreds of contact sport athletes and others through postmortem examination, and there is a strong causal relationship between repetitive head impacts (RHI) and CTE. The neuropathological features of CTE are unique, and the NINDS-NIBIB criteria have greatly advanced the diagnosis of CTE.

Summary: Traumatic brain injury is a major cause of death worldwide, leading to cellular physiological changes, neuronal cell death, altered molecular pathways, and decreased quality of life. The use of alternative animal models that better replicate human individuality, such as those with evolved brain protection, may provide preventive applications for humans. Studying these unconventional animals can increase knowledge, improve current animal models, and inspire translational applications in human brain injury research.

Summary: Tauopathies are neurodegenerative diseases characterized by abnormal tau protein-containing neurofibrillary tangles (NFTs). This study utilized an AI-based method to quantitatively analyze tau pathology and found that AI-derived NFT metrics significantly predicted cognitive impairment in PART patients.

Summary: Pathological tau aggregation is a key feature of many neurodegenerative diseases, but varies widely in affected brain regions, symptoms, morphology, conformation, and isoform ratio. Tau interactome studies provide important insights into disease mechanisms and cellular environment during tau aggregation. Analysis of 12 tau interactome studies revealed consistent interactions with proteins involved in RNA binding, ribosome, and proteasome function, with differences observed between human and rodent studies. Dysregulation of tau interactions was observed during the development of pathology, suggesting a role for RNA binding proteins in driving tau pathology.

Summary: This study aimed to explore the relationship between the neuropsychological profile and traumatic load in American football players, whether traumatic brain injury has a specific cerebral perfusion pattern, and whether this pattern correlates with neuropsychological performances. The results found that cognitive performances of American football players were associated with field position and career length, with linemen having poorer executive, verbal, and visual performances. Additionally, American football players had reduced cerebral blood flow values in sensory-motor areas, and poorer neuropsychological performances correlated with lower perfusion in specific brain areas.

Summary: This study found that neuronal p-tau was significantly higher than astrocytic p-tau in various cortical regions. Astrocytic p-tau in the sulcus was primarily localized to the subpial region as thorn-shaped astrocytes, a form of age-related tau astrogliopathy. Neuronal p-tau was significantly associated with age, years of RHI exposure, and CTE severity, while astrocytic p-tau pathology was only significantly associated with age.

Summary: This study describes the clinical and genetic findings of a patient with pure FTD who carries the p.Asp395Gly VCP variant, which is associated with familial FTD with tauopathy. The patient exhibited self-centered behavior, executive dysfunction, and bilateral frontal lobe atrophy. The absence of comorbid muscle or bone disease suggests that VCP pathogenic variants should be considered in sporadic FTD cases as well. CSF analysis indicated FTD due to NFT accumulation.

Summary: Traumatic brain injury (TBI) causes immediate tissue and cellular damage to the brain, followed by secondary damage that can lead to chronic neurodegeneration and cognitive deficits. TBI is also associated with a higher risk of developing neurodegenerative diseases such as Alzheimer's disease, frontotemporal dementia, and amyotrophic lateral sclerosis. However, the prevalence of TDP-43 pathology after TBI has not been fully studied.
