Notch signaling in leukemias and lymphomas

Aberrant Notch activation is linked to cancer since 1991 when mammalian Notch1 was first identified as part of the translocation t(7; 9) in a subset of human T-cell acute lymphoblastic leukemias (T-ALL). Since then oncogenic Notch signaling has been found in many solid and hematopoietic neoplasms. Depending on tumor type Notch interferes with differentiation, proliferation, survival, cell-cycle progression, angiogenesis, and possibly self-renewal. In hematopoietic neoplasms, recent findings indicate an important role of Notch for TALL induction and progression and the pathogenesis of human T- and B-cell-derived lymphomas. Notch signaling has been identified as a potential new therapeutic target in these hematopoietic neoplasms. This review will focus on the most recent findings on Notch signaling in leukemias and lymphomas and its potential role in the maintenance of malignant stem cells.

Summary: This study found that mechanical loading can enhance bone mass, prevent osteolytic destruction, and inhibit the growth and dissemination of multiple myeloma cells in mice. These findings suggest that physical stimuli may serve as an anabolic treatment for osteolytic bone destruction in patients with multiple myeloma.

Summary: This study investigated the bacterial composition of scalp skin and hair follicles in patients with FFA, LPP, AAc, and healthy individuals, finding dysbiosis in the bacterial communities of FFA and LPP patients and observed molecular signals associated with increased expression in lesional sites. This suggests the need for further research on the role of exogenous factors and molecular interactions in disease onset and propagation.

Prevention of Bone Destruction by Mechanical Loading Is Not Enhanced by the Bruton's Tyrosine Kinase Inhibitor CC-292 in Myeloma Bone Disease

Summary: Research showed that the Bruton's tyrosine kinase inhibitor CC-292 did not significantly enhance bone anabolism or rescue bone mass compared to mechanical loading alone in the treatment of myeloma bone disease. Novel anti-myeloma and anti-resorptive strategies in combination with physical stimuli are needed to improve the treatment of MMBD.

Estimating infectiousness throughout SARS-CoV-2 infection course

Summary: In a study of SARS-CoV-2 cases in Germany, it was found that younger subjects had lower viral loads but higher cell culture replication probabilities compared to older subjects. Additionally, 8% of subjects had viral loads above 10(9) copies per swab, with a third of them being presymptomatic, asymptomatic, or mildly symptomatic individuals with a mean age of 37.6 years. The B.1.1.7 lineage infections had a higher viral load and cell culture replication probability than non-B.1.1.7 subjects.

Summary: This study investigated the effects of different cosmetic lotions on skin microbiome and skin physiology after mild tape stripping. The results showed that the lotions accelerated the recovery of the microbiome and increased stratum corneum hydration. The composition of the lotion and induced repair mechanism may be responsible for the accelerated normalization of the microbiome.

Sonication versus the conventional method for evaluation of the dental microbiome: a prospective pilot study

Summary: This study investigated the use of sonication as a new tool in microbiological probing of dental infections. The results showed that sonication yielded a higher number of bacterial sequencing reads and identified more operational taxonomic units (OTUs) compared to the standard procedures. It also found that actinomycetales were more abundant in sonication fluid samples. Additionally, due to the high rates of antibiotic resistance, a more targeted antibiotic therapy is favored, with levofloxacin being a potential alternative to amoxicillin + clavulanate in patients with penicillin allergy.

Summary: Studies on anagen hair follicles have identified the presence of bacteria in the lower follicles, with Lawsonella clevelandensis, Staphylococcaceae, and Propionibacteriaceae being the most abundant. These findings suggest that the skin microbiome may play a role in hair cycling through its influence on the local immune system, particularly in relation to bacterial presence near follicular immune-privileged areas.
