Summary: Liver disease is a significant clinical issue affecting 600 million people worldwide. Liver transplantation, although the gold standard treatment for liver failure, currently meets less than 10% of the global demand due to a shortage of available organs. Cell- and tissue-based therapies offer a promising alternative to organ transplantation.

Summary: Diabetes is a metabolic disease characterized by insulin deficiency. Bioengineering of stem cells to restore insulin production has the potential to cure diabetes. This review focuses on the recent developments in bioengineering iPSCs, MSCs, ESCs, and pancreatic progenitor cells to generate insulin producing and glucose regulating cells.

Summary: Volumetric muscle loss (VML) is an irreversible loss of skeletal muscle due to trauma or surgery. In this study, a macroporous dECM-methacrylate (dECM-MA) hydrogel was developed and found to promote cellularization, endothelialization, and establishment of a pro-regenerative immune microenvironment. The hydrogel also enhanced the proliferation of transplanted primary muscle stem cells, muscle tissue regeneration, and functional recovery.

Summary: Cardiovascular disease is the leading cause of adult death worldwide, with myocardial infarction accounting for four fifths of cardiovascular disease deaths. Regenerative medicine, particularly using stem cells to repair the heart, has shown promising benefits as a treatment for ischemic heart disease. Despite the potential of stem cell therapy, more research is needed to fully understand its mechanism of action and its long-term effects on cardiovascular disease.

Summary: Osteoporosis is a systemic metabolic bone disease characterized by bone loss and microstructural degeneration, leading to increasing personal and societal costs. MSC transplantation shows promising potential in treating osteoporosis, but issues such as safety, transplant efficiency, and standardization of the manufacturing process need to be addressed. Comprehensive evaluation of clinical trials is essential for clinical translation of MSC therapy for osteoporosis.

Summary: The pathogenesis of sepsis is an imbalance between pro-inflammatory and anti-inflammatory responses. MSC-derived extracellular vesicles (EVs) have shown beneficial effects for the treatment of acute lung injury (ALI) and sepsis. However, the present study demonstrated that EVs isolated from bone marrow-derived MSCs failed to attenuate the severity of multiorgan dysfunctions.

Summary: Implanting MSCs in SCI repair holds promise, but the harsh environment and excessive ROS threaten MSC survival. The dual-phase pathology of acute and chronic phases makes it challenging to alleviate severe inflammation and promote functional recovery. The RF-pGel-MS scaffold system efficiently scavenges ROS and boosts stem cell therapeutics, showing significant efficacy in locomotor and electro-physiology recovery in SCI rats.
