High-density lipoproteins (HDL) are classified as atheroprotective because they are involved in transport of cholesterol to the liver, known as reverse cholesterol transport (RCT) exerting antioxidant and anti-inflammatory activities. There is also evidence for cytoprotective, vasodilatory, antithrombotic, and anti-infectious activities for these lipoproteins. HDLs are known by structural, metabolic and biologic heterogeneity. Thus, different methods are able to distinguish several subclasses of HDL. Different separation techniques appear to support different HDL fractions as being atheroprotective or related with lower cardiovascular (CV) risk. However, HDL particles are not always protective. Modification of constituents of HDL particles (primarily in proteins and lipids) can lead to the decrease in their activity and induce proatherogenic properties, especially when isolated from patients with augmented systemic inflammation. According to available studies, it seems that HDL functionality may be a better therapeutic target than HDL cholesterol quantity; however, it is still disputable which subfractions are most beneficial. There is mounting evidence supporting HDL subclasses as an important biomarker to predict and/or reduce CV risk. In this review we discuss recent notices on atheroprotective and functional characteristic of different HDL subfractions. Also, we provide a brief overview of the different methods used by clinicians and researchers to separate HDL subfractions. Ongoing and future investigations will yield important new information if any given separation method might represent a 'gold standard', and which subfractions are reliable markers of CV risk and/or potential targets of novel, more focused, and effective therapies.

Non-alcoholic fatty liver disease, metabolic syndrome, and type 2 diabetes mellitus: where do we stand today?

Bojan Mitrovic, Zoran M. Gluvic, Milan Obradovic, Maja Radunovic, Manfredi Rizzo, Maciej Banach, Esma R. Isenovic

Summary: Non-alcoholic fatty liver disease (NAFLD), metabolic syndrome (MetS), and type 2 diabetes (T2DM) are highly prevalent metabolic disorders with significant impacts on global public health. MetS is a complex condition characterized by glucose intolerance, insulin resistance, dyslipidemia, inflammation, and hypertension. NAFLD, recognized as a hepatic manifestation of MetS, is often associated with T2DM. This review examines the underlying connections between these diseases and the risks associated with their co-occurrence.

The Immunogenic Potential of PCSK9 Peptide Vaccine in Mice

Sarina Ataei, Amir Abbas Momtazi-Borojeni, Shiva Ganjali, Maciej Banach, Amirhossein Sahebkar

Summary: This study evaluated the immunogenic potential of a carrier-free peptide-based anti-PCSK9 vaccine in albino mice. The results showed that the vaccine induced a significant production of IgG antibodies and inhibited PCSK9 concentrations and its interaction with LDLR in the blood plasma.

The effects of statins on the function and differentiation of blood cells

Efficacy and safety of sacubitril/valsartan in heart failure compared to renin-angiotensin-aldosterone system inhibitors: a systematic review and meta-analysis of randomised controlled trials

Adrian V. Hernandez, Vinay Pasupuleti, Nancy Scarpelli, Jack Malespini, Maciej Banach, Agata M. Bielecka-Dabrowa

Summary: This study assessed the benefits and harms of Sacubitril/valsartan (S/V) compared to ACEI or ARB in heart failure patients. The results showed that S/V reduced HF hospitalizations by 20%, CV mortality by 14%, and all-cause mortality by 11% compared to ACEI or ARB. However, S/V had negative effects on renal function and increased hypotension events.

Effects of statins on the incidence and outcomes of acute kidney injury in critically ill patients: a systematic review and meta-analysis

Summary: AKI is a common and severe complication in critically ill patients, with high mortality rates. Statin therapy may have beneficial effects on the disease by reducing mortality rates and overall hospital length of stay. However, the impact of statin treatment on the development of AKI and ICU stay is inconclusive.

Age-Related Differences in the Role of Risk Factors for Ischemic Stroke

George Howard, Maciej Banach, Brett Kissela, Mary Cushman, Paul Muntner, Suzanne E. Judd, Virginia J. Howard

Summary: This study assessed the association between stroke risk factors and incident stroke, and evaluated whether the magnitude of this association is uniform across different age groups. The results showed that the associations of hypertension and diabetes with stroke significantly decreased in older age groups, while the associations of smoking, atrial fibrillation, and left ventricular hypertrophy with stroke did not vary with age.

Summary: Pre-obesity is a condition that increases the risk of obesity, cardiovascular diseases, and diabetes. A study found that Altilix(R), a nutraceutical containing chlorogenic acid and luteolin extracts, can improve hepatic and cardio-metabolic parameters. In a post-hoc analysis, Altilix(R) was shown to effectively improve anthropometry, glucose metabolism, lipid profile, hepatic functionality, carotid-media thickness, and endothelial function in pre-obesity subjects.

On the Need for a Paradigm Change in the Valuation of Concrete with Waste Materials Based on the Example of Concrete with Crumb Rubber

Julia Nowak, Roman Jaskulski, Wojciech Kubissa, Bartlomiej Matusiak, Maciej Banach

Summary: This study investigated the strength and thermal properties of concrete with crumb rubber from used tyres replacing part of the aggregate. The results showed a decrease in compressive strength and improved insulation properties with an increase in rubber content. A thermomechanical index was introduced to evaluate the functionality of the concrete, and only three out of twelve series failed to meet the performance requirements.

Summary: The COVID-19 pandemic, which started in Wuhan, China, in early 2020, has caused a global health crisis. The disease mainly affects the respiratory system but can also have an impact on other systems, including the cardiovascular system. The relationship between atherosclerosis and COVID-19 is bidirectional, with both conditions exacerbating each other. The pandemic has also led to reduced access to healthcare amenities, an increase in sedentary lifestyles and unhealthy eating habits, resulting in a high prevalence of overweight and obese individuals. The long-term healthcare challenges posed by COVID-19 can be mitigated by the lessons learned and the advancements made in patient care during this crisis.

Does Iodine Influence the Metabolism of Glucose?

Ioannis Ilias, Charalampos Milionis, Lina Zabuliene, Manfredi Rizzo

Summary: Thyroid function and glucose status are closely linked, and studies have shown the relationship between iodine and glucose metabolism, diabetes, and prediabetic conditions. The potential beneficial effect of iodine on glucose metabolism may be due to its antioxidant properties. However, further research is needed to fully understand the mechanism behind this relationship.
