This review focuses on disease-associated germline coding mutations in the aldo-keto reductase (AKR) superfamily, uncovering the effects of these mutations on androgen metabolism, bile acid deficiency, and other diseases. However, further research is needed to fully understand the relationship between AKR mutations and diseases.

Summary: Anthracyclines are highly effective in cancer treatment, but their cardiotoxicity limits their prescribed dosage due to the risk of heart failure. The cardiotoxicity is likely caused by the generation of reactive oxygen species (ROS) or alcohol metabolites from reduction reactions in the structure of anthracyclines. ROS is responsible for acute cardiotoxicity, while alcohol metabolites contribute to progressive cardiomyopathy and congestive heart failure. This review explores the molecular pathways of anthracycline-induced cardiotoxicity, with a focus on the role of alcohol metabolites and ROS. It also discusses effective strategies to minimize or terminate cardiotoxicity.

Summary: A copper(I)-catalyzed asymmetric 1,3-dipolar cycloaddition of azomethine ylides and 1,3-enynes is reported, providing chiral poly-substituted pyrrolidines with high selectivities. 4-aryl-1,3-enynes and 4-silyl-1,3-enynes are suitable dipolarophiles, while 4-alkyl-1,3-enynes are inert. The method is capable of constructing tetrasubstituted stereogenic carbon centers and chiral spiro pyrrolidines. DFT calculations suggest a concerted mechanism. Transformations starting from the pyrrolidine bearing a triethylsilylethynyl group and centered on the alkyne group are achieved.

Summary: Irisin is a cytokine secreted by myocytes in response to exercise, and its levels can be measured in blood. However, blood collection is invasive, and saliva samples could provide a less invasive alternative for irisin detection. In this research, serum and saliva samples were collected from healthy male adults before and after exercise, and it was found that both serum and salivary irisin levels increased at 24 hours post exercise and returned to baseline after 48 hours. The study suggests that saliva samples could be a valid and less invasive method for detecting exercise-induced changes in irisin levels.

Summary: The role of oxidative stress (OS) in male infertility is well established and it can also be a factor in other conditions such as inflammation, varicocele, and gonadotoxin effects. Reactive oxygen species (ROS) play important roles in spermatogenesis and fertilization, and recent studies have identified epigenetic mechanisms that can be inherited by offspring. When ROS production is excessive, it leads to OS and subsequent damage to lipids, proteins, and DNA, resulting in infertility and/or premature pregnancy termination. Understanding the delicate balance between ROS and antioxidants, as well as measuring total antioxidant capacity (TAC) in seminal plasma, is important for personalized approaches to male infertility treatment.
