Metabolism of Plant Polyphenols in the Skin: Beneficial Versus Deleterious Effects

Polyphenols are produced by all higher plants in order to protect them against biotic and abiotic stress such as UV radiation, temperature changes, infections, wounding, and herbivores. When in contact with human skin, polyphenols exert either curative or damaging action depending on their physical-chemical properties, bioavailability through cutaneous barrier, metabolism in the skin, and individual sensitivity. This review will focus on 1) synthesis and metabolism of polyphenols and their role in the plant physiology, 2) non-enzymatic and enzymatic polyphenol transformation in the skin, 3) polyphenols as inhibitors or inducers of inflammatory response in the skin, and 4) photo-protective versus photo-toxic effects of polyphenols. The potential consequences of these controversial effects on the use of plant polyphenols in dermatology and cosmetology will be also discussed.

Summary: This study investigated the effects of plant polyphenolic compounds (PPs) on the responses of human HaCaT keratinocytes to UV-C radiation. The results showed that PPs added immediately after UV-C exposure had a cytoprotective effect, with acacetin being the most effective. Furthermore, PPs were found to reduce DNA damage in the nuclei of keratinocytes exposed to UV-C.

Effects of native and particulate polyphenols on DNA damage and cell viability after UV-C exposure

Summary: Due to poor water solubility of plant polyphenols, their bioavailability is low. To overcome this limitation, polymeric materials can be used to coat drug molecules with multiple layers. Microcrystals of quercetin and resveratrol coated with (PAH/PSS)(4) or (CH/DexS)(4) shell were prepared. UV-C induced damage to human HaCaT keratinocytes was evaluated by treating cells with native and particulate polyphenols. The results showed that both native and particulate polyphenols increased cell viability after UV-C exposure in a dose-dependent manner, with particulate quercetin showing a more pronounced effect. Quercetin coated with (CH/DexS)(4) shell effectively reduced DNA lesions in keratinocyte nuclei, while both native and particulate resveratrol were ineffective against DNA damage. Quercetin enhances cell survival and DNA repair capacity, with the coating of (CH/DexS)(4) shell further enhancing its impact on DNA repair.

Summary: Research on ultraweak photon emission (UPE) in living organisms dates back to the 1920s, with the discovery of mitogenetic radiation (MGR) by Russian biologist A. Gurwitsch. While there have been numerous studies confirming the existence of permanent biophoton emission and intercellular electromagnetic communication, the significance of MGR has been a topic of debate. New evidence suggests that non-chemical signals can trigger biological cellular responses in a synchronous manner.
