Summary: In this study, a multifunctional polysaccharide nanoplatform called GH-PID was constructed for the treatment of hepatocellular carcinoma (HCC). The GH-PID nanodonuts showed excellent HCC-targeted ability and photochemotherapy effect. The nanodonuts also induced immunogenic cell death (ICD) and enhanced antitumor immunity, leading to the eradication of primary tumors and inhibition of distant tumor growth and lung metastasis in HCC mouse models.

Summary: This study investigates the role of mitochondrial translocator protein (TSPO) in the regulation of ferroptosis and antitumor immunity in hepatocellular carcinoma (HCC). The results show that TSPO promotes HCC cell growth, migration, and invasion, and inhibits ferroptosis through enhancing the Nrf2-dependent antioxidant defense system. Furthermore, TSPO upregulates PD-L1 expression and promotes HCC immune escape. Targeting TSPO can be a promising new strategy for HCC treatment.

Summary: N-acetylaspartate (NAA) is synthesized by the mitochondrial enzyme NAT8L using acetyl-CoA and aspartate as substrates. The modulation of NAT8L expression has been found to affect the metabolic adaptations and proliferative capability of hepatocellular carcinoma cells. NAT8L downregulation increases cell proliferation, while over-expression decreases cell growth. Mechanistically, NAT8L downregulation promotes aspartate outflow from mitochondria to enhance glucose flux into the pentose phosphate pathway, resulting in increased purine biosynthesis. These findings suggest that NAT8L plays a critical role in regulating cell proliferation through metabolic rewiring.

Summary: Incomplete radiofrequency ablation (iRFA) leads to the survival of cells that contribute to hepatocellular carcinoma (HCC) recurrence. This study investigated the role of reactive oxygen species (ROS) in HCC cells exposed to sublethal heat stress. It was found that sublethal heat stress induced a burst of ROS, which was initiated by upregulated NOX4 expression in mitochondria. The increased ROS triggered PINK1-dependent mitophagy to protect cells from apoptosis.

Summary: Obesity and hypercholesterolemia are major risk factors for hepatocellular carcinoma. This study found that obesity leads to hepatic cholesterol accumulation, which suppresses natural killer T (NKT) cell-mediated antitumor immunosurveillance. By lowering cholesterol and inhibiting cholesterol biosynthesis, the function of NKT cells can be restored to prevent HCC development promoted by obesity.
