Function of the pseudo phosphotransfer proteins has diverged between rice and Arabidopsis

John Vaughan-Hirsch, Emily J. Tallerday, Christian A. Burr, Charlie Hodgens, Samantha L. Boeshore, Kevin Beaver, Allison Melling, Kartika Sari, Ian D. Kerr, Jan Simura, Karin Ljung, Dawei Xu, Wanqi Liang, Rahul Bhosale, G. Eric Schaller, Anthony Bishopp, Joseph J. Kieber

Summary: The rice PHP genes have been identified as negative regulators of cytokinin signaling, affecting multiple aspects of plant growth and development. However, unlike Arabidopsis, disruption of the rice PHPs does not affect root vascular patterning.

Preparation, Scanning and Analysis of Duckweed Using X-Ray Computed Microtomography

Dylan H. Jones, Brian S. Atkinson, Alexander Ware, Craig J. Sturrock, Anthony Bishopp, Darren M. Wells

Summary: This study introduces a new sample handling protocol for mu CT imaging of aquatic plant material, using duckweed plants and turions as exemplars, and compares it with existing approaches. The technique enables previously unseen 3D volume analysis of gaseous filled spaces, cell material, and sub-cellular features, providing novel insights for anatomical structural analysis.

Systems approaches reveal that ABCB and PIN proteins mediate co-dependent auxin efflux

Nathan L. Mellor, Ute Voss, Alexander Ware, George Janes, Duncan Barrack, Anthony Bishopp, Malcolm J. Bennett, Markus Geisler, Darren M. Wells, Leah R. Band

Summary: Members of the B family of membrane-bound ATP-binding cassette (ABC) transporters play important roles in auxin efflux in plants. This study integrates ABCB localization data into a multicellular model to predict the impact of ABCB-mediated auxin transport on organ-scale distribution. The results suggest that ABCBs enable auxin efflux independently of PINs, but PINs predominantly mediate auxin efflux when co-localized with ABCBs.

Transcriptional reprogramming during floral fate acquisition

Summary: This study reveals the transcriptional network induced by auxin at the early stage of flower development and suggests that auxin initiates growth through antagonistic regulation of growth-promoting and growth-repressive hormones, quasi-synchronously to floral fate specification. Additionally, two DNA-binding One Zinc Finger (DOF) transcription factors acting in an auxin-dependent network were identified to interface growth and cell fate from the early stages of flower development onward.

Putative pectate lyase PLL12 and callose deposition through polar CALS7 are necessary for long-distance phloem transport in Arabidopsis

Lothar Kalmbach, Matthieu Bourdon, Ilya Belevich, Jerome Pelloux, Eija Jokitalo, Josip Safran, Adrien Lemaire, Jung-ok Heo, Sofia Otero, Bernhard Blob, Yka Helariutta

Summary: In plants, the phloem relies on specialized cells called sieve elements for long-distance distribution of photosynthetic products. This study identifies PLL12 as a critical gene for sieve element differentiation and suggests its vital role in pectin remodeling. While short distance symplastic diffusion is unaffected, PLL12 appears to act as a regulator for local pectin degradation during sieve pore maturation.

In planta dynamics, transport biases, and endogenous functions of mobile siRNAs in Arabidopsis

Emanuel A. Devers, Christopher A. Brosnan, Alexis Sarazin, Gregory Schott, Peiqi Lim, Satu Lehesranta, Yrjo Helariutta, Olivier Voinnet

Summary: The mechanism of RNA interference (RNAi) in plants is not well understood, especially the propagation and selectivity of siRNAs. Through research, it has been found that RNAi in plants is propagated through plasmodesmata (PDs) and involves specific ARGONAUTE (AGO) proteins. Stress enhancement also plays a role in the translocation and regulation of endogenous siRNAs.

Summary: Research has found that m6A modification can affect auxin-related defects and auxin signaling in Arabidopsis thaliana mutants, as well as lead to defects in endodermal patterning in the primary root.

Summary: This study provides a detailed analysis of the phloem poles using a second-generation single-cell approach, revealing the similarities in gene regulatory networks and developmental trajectories associated with phloem development. Additionally, a set of key transcription factors that play a crucial role in the early development of the phloem have been identified.

Summary: The TMO5/LHW complex plays a crucial role in regulating cytokinin levels in the root meristem, influencing vascular development. Xylem cells act as a local organizer of vascular development, non-autonomously regulating cytokinin levels in neighbouring procambium cells.
