Angioprevention with fenretinide: Targeting angiogenesis in prevention and therapeutic strategies

Clinical trials have revealed that N-(4-hydroxyphenyl) retinamide (4HPR; fenretinide), a synthetic retinoic acid derivative, is a highly active and promising therapeutic and chemopreventive agent. Fenretinide shows biological activity against numerous cancer types in vitro and in preclinical studies. Clinical trials have shown that fenretinide induces a significant reduction of second breast cancer in premenopausal women. Several studies on different neoplasms are ongoing, such as breast and ovarian cancer, neuroblastoma, glioblastoma, head and neck and skin cancers and others. It has minimal side effects in humans, so that trials in young women at high-risk of breast cancer and ovarian and for the prevention of other tumor types such as lung cancer could be envisaged. Here we review some ongoing clinical trials and evaluate the possible mechanisms underlying the secondary chemopreventive effects of 4HPR. In particular we report basic and translational data on the anti-angiogenic angiopreventive properties of fenretinide, its anti-invasive activity, its ability to induce apoptosis and to generate or enhance production of reactive oxygen species as possible molecular bases for a chemopreventive action in patients. Summary: Immune checkpoint inhibitors have been widely used in the treatment of non-small cell lung cancer, but not all patients benefit from them. Other cells in the tumor microenvironment, such as NK cells, also play a role and may affect the response to immune checkpoint inhibitors. Further research is needed to understand the role of pro-inflammatory NK cells in NSCLC and their impact on treatment response to ICIs.

Human RNASET2: A Highly Pleiotropic and Evolutionary Conserved Tumor Suppressor Gene Involved in the Control of Ovarian Cancer Pathogenesis

Summary: Ovarian cancer is a malignant gynecological cancer with a 5-year survival rate of 25-30% in the last decade. Cancer immunotherapy is considered a promising strategy for ovarian cancer treatment. The human RNASET2 gene plays a crucial role in suppressing tumor diversity by establishing functional communication between ovarian cancer cells and innate immune system effectors. The protein product of RNASET2 promotes the recruitment of anti-tumoral macrophages to the cancer tissue. This finding suggests the potential of developing a multitasking therapeutic approach for ovarian cancer treatment.

The Microalgal Diatoxanthin Inflects the Cytokine Storm in SARS-CoV-2 Stimulated ACE2 Overexpressing Lung Cells

Summary: This study demonstrates the anti-inflammatory effects of Diatoxanthin (Dt) on human lung cells, specifically the A549-hACE2 cell line. Dt enhances cell metabolism and ACE2 enzymatic activity, while decreasing the production of interleukin-6 and increasing the release of interleukin-10 in response to the SARS-CoV-2 spike glycoprotein. Dt also upregulates genes and proteins related to the interferon pathway and innate immunity response. These findings suggest that Dt may be a promising therapeutic agent for the treatment and/or prevention of severe inflammatory syndrome associated with SARS-CoV-2 infection.

Taste and Smell Disorders in Cancer Treatment: Results from an Integrative Rapid Systematic Review

Summary: Taste and smell disorders are common side effects in cancer patients, and identifying the treatments that cause them is important for improving patients' quality of life. This review examined the oncological treatments associated with taste and smell changes and their timing of onset. Fourteen studies were included, showing a high heterogeneity. Drugs such as Docetaxel, paclitaxel, nab-paclitaxel, capecitabine, cyclophosphamide, epirubicin, anthracyclines, and oral 5-FU analogues were found to be the most frequently associated with taste and smell disorders. The review highlights the need for further research, particularly focused on prevention.

Summary: The exploration of natural preventive molecules from marine microorganisms for nutraceutical and pharmaceutical use has increased. In this study, an extract from the marine diatom Skeletonema marinoi and its carotenoid pigment fucoxanthin showed potential chemopreventive and anti-angiogenic activities against prostate cancer and endothelial cells. The extract and fucoxanthin inhibited cell growth and hindered vascular network formation. Molecular analysis revealed modulations in angiogenesis and inflammation molecules. These findings highlight the potential of microalgae-derived drugs as a source of novel nutraceuticals for cancer prevention.

Novel Treatments for Obesity: Implications for Cancer Prevention and Treatment

Summary: It is established that obesity is associated with higher cancer incidence. Effective obesity treatment provides new perspectives in managing a modifiable cancer risk factor. This review summarizes the correlations between weight loss in obesity and cancer, exploring the potential of obesity treatment in clinical cancer management. The evidence for the effects of obesity therapy on proliferation, apoptosis, and chemotherapy response is reviewed. Further studies and collaboration among oncologists and obesity treatment experts are needed to fully evaluate the relationship between anti-obesity treatment and cancer.
