Summary: The endoplasmic reticulum (ER) is the primary storage compartment for calcium (Ca2+) in eukaryotic cells. Ca2+ is mobilized from the ER through store-operated Ca2+ channels (SOCCs), leading to a store-operated Ca2+ entry (SOCE) that has been extensively studied in non-excitable cells. The presence of this Ca2+ entry route in neurons has been a subject of debate, but experimental evidence suggests that SOCE can be generated by the recruitment of Ca2+ from neuronal ER Ca2+ stores. This review summarizes the main supporting studies and explores the molecular composition, expression, pharmacological properties, and physiological relevance of neuronal SOCCs.

Summary: Research shows that TRPC6 channels play a crucial role in podocyte function and their dysregulation is associated with kidney diseases. Through single channel patch clamp technique, it is found that TRPC6 channels in human podocytes and rat glomerular podocytes are sensitive to calcium store depletion. In a rat model of DM2, a reduction in store-operated calcium entry was observed in glomerular podocytes, along with a reorganization of calcium influx mediated by TRPC6 channels and ORAI.

Summary: Ca2+ signaling is crucial for T lymphocyte activation, an important step in autoimmune diseases like multiple sclerosis. Store-operated Ca2+ entry (SOCE) plays a vital role in T lymphocyte activation, including the nuclear localization of NFAT. In this study, the synthetic compound STX140 was found to inhibit SOCE in T lymphocytes and decrease the secretion of pro-inflammatory cytokines. Moreover, STX140 completely blocked clinical symptoms in an animal model of MS, showing potential as a new therapeutic option.

Summary: The natural compound 3,3'-Diindolylmethane (DIM) inhibits proliferation and induces apoptosis and autophagy in gastric cancer cells by activating the STIM1-mediated Store-operated calcium entry (SOCE) pathway. Inhibition of SOCE attenuates DIM-induced effects on apoptosis and autophagy, suggesting SOCE as a potential therapeutic target in gastric cancer. Further research is warranted to explore combination chemotherapy strategies targeting SOCE for clinical applications.

Summary: This study evaluated the recellularization of blood vessel conduits of acellular lung scaffolds with different types of endothelial cells, showing that RMEPCs have the capability to repopulate all vascular segments, while PAECs and MVECs exhibit selective tropism for larger vessels or microvasculature, respectively. The RMEPC phenotype described in the study could be valuable for identifying suitable endothelial progenitors for efficient vascular organ and tissue engineering, regeneration, and repair.
