Investigating the possible causal association of coffee consumption with osteoarthritis risk using a Mendelian randomization analysis

This study examined whether coffee consumption is causally associated with osteoarthritis. A two-sample Mendelian randomization (MR) analysis using inverse-variance weighted (IVW) and weighted median estimates, and the MR-Egger regression method were performed. The publicly available summary statistical datasets of coffee consumption genome-wide association studies (GWASs) meta-analyses on coffee intake from eight Caucasian cohorts (n=18,176), GWAS meta-analyses of predominately regular-type coffee consumers of European ancestry (n = up to 91,462), and a GWAS in 7410 patients with osteoarthritis in the arcOGEN study with 11,009 controls of European ancestry were evaluated. Four single-nucleotide polymorphisms (SNPs) from GWASs of coffee consumption as instrumental variables (IVs) to improve inference were selected. These SNPs were located at neurocalcin delta (NCALD) (rs16868941), cytochrome p450 oxidoreductase (POR) (rs17685), cytochrome p450 family 1 subfamily A member 1 (CYP1A1) (rs2470893), and neuronal cell adhesion molecule (NRCAM) (rs382140). The IVW method (beta=0.381, SE=0.170, p=0.025) and the weighted median approach (beta=0.419, SE=0.206, p=0.047) showed evidence to support a causal association between coffee consumption and osteoarthritis. MR-Egger regression revealed that directional pleiotropy was unlikely to be biasing the result (intercept=0.064; p=0.549), but showed no causal association between coffee consumption and osteoarthritis (beta=-0.518, SE=1.270, p=0.723). Cochran's Q test and the funnel plot indicated no evidence of heterogeneity between IV estimates based on the individual variants. The results of MR analysis support the observation that coffee consumption is causally associated with an increased risk of osteoarthritis.

Summary: This meta-analysis compared the effectiveness and safety of TNFI biosimilars to TNFI originators in RA patients. The results showed that TNFI biosimilars had comparable efficacy to their originators in terms of ACR20 and ACR50 response rates. There was also no significant difference in adverse events between the two groups.

Comparative efficacy and safety of infliximab and its biosimilars in patients with rheumatoid arthritis presenting an insufficient response to methotrexate A network meta-analysis

Summary: This study aimed to compare the relative efficacy and safety of infliximab and its biosimilars in patients with active rheumatoid arthritis (RA) who showed an inadequate response to methotrexate (MTX). A Bayesian network meta-analysis was conducted, combining direct and indirect evidence from randomized controlled trials (RCTs) to compare the efficacy and safety of infliximab biosimilars and the originator product in this patient population. The results showed that there were no significant differences in ACR20 response rates and serious adverse events (SAEs) between infliximab biosimilars and the originator.

A network meta-analysis of randomized controlled trials comparing the effectiveness and safety of voclosporin or tacrolimus plus mycophenolate mofetil as induction treatment for lupus nephritis

Summary: This study compared the effectiveness and safety of voclosporin + MMF, tacrolimus + MMF, and monotherapy for induction treatment in lupus nephritis patients. The results showed that tacrolimus + MMF and voclosporin + MMF were more effective than monotherapy, with tacrolimus + MMF being the most effective. However, tacrolimus + MMF had a higher risk of serious adverse events compared to monotherapy.

Efficacy and safety of tacrolimus versus mycophenolate mofetil as induction treatment and low-dose tacrolimus as treatment for lupus nephritis: a meta-analysis

Summary: This study compared the efficacy and safety of tacrolimus and mycophenolate mofetil (MMF) as induction therapy for lupus nephritis (LN). The results showed that there was no difference between tacrolimus and MMF in terms of complete or partial remission rates. However, tacrolimus had a lower incidence of herpes zoster infection and a higher risk of serum creatinine elevation compared to MMF. Overall, 3 mg/d tacrolimus was proven to be an effective and safe treatment for LN.

Comparison of the efficacy and safety of tocilizumab, sarilumab, and olokizumab in patients with active rheumatoid arthritis: a network meta-analysis of randomized controlled trials

Summary: This study compared the efficacy and safety of olokizumab, tocilizumab, and sarilumab in rheumatoid arthritis patients who were intolerant or inadequately responding to methotrexate. The results showed that tocilizumab had the highest ACR20 response rate, followed by sarilumab and olokizumab. These three drugs were more effective than adalimumab and had similar safety profiles.

Circulating leptin level in osteoarthritis and associations between leptin receptor polymorphisms and disease susceptibility: A meta-analysis

Summary: This study aimed to systematically review the relationship between circulating leptin levels and osteoarthritis (OA), as well as the association between leptin receptor (LEPR) polymorphisms and OA susceptibility. Meta-analysis results showed that patients with OA had significantly higher levels of circulating leptin and synovial leptin compared to healthy individuals. Additionally, the LEPR rs1137101 polymorphism was associated with an increased risk of developing OA.

Association between the functional FCGR3A F158V and FCGR2A R131H polymorphisms and responsiveness to biologics in rheumatoid arthritis patients: A meta-analysis

Summary: This study investigated the association between FCGR3A V158F and FCGR2A R131H polymorphisms and the responsiveness to biologic therapy in RA patients. The study found that the FCGR3A V allele was associated with better responsiveness to rituximab, while FCGR2A R allele carriers may show a better response to biologics. Genotyping of these polymorphisms could be useful in predicting the responsiveness to biologic treatment in RA patients.

Association between Susceptibility to Systemic Lupus Erythematous and Polymorphisms in miR-146a and miR-499: A Meta-Analysis

The results showed that the miR-146a rs2431697 C allele is associated with SLE and has a protective effect. However, the miR-146a rs2910164 is not associated with susceptibility to SLE.
