Summary: In this study, a series of azomethine derivatives were synthesized and their anti-inflammatory potency was investigated. Computational docking studies showed better affinity of the compounds towards COX-2 compared to COX-1, with compounds 4a, 4d, and 4k exhibiting the best binding. In vitro inhibition assays confirmed that the synthesized compounds had less inhibition on COX-1 enzyme than COX-2 enzyme. Compound 4d showed the highest inhibition of inflammation in in-vivo screening. The compounds also demonstrated good antioxidant properties but minimal cytotoxicity on cancer cells.

Summary: Two novel series of compounds derived from nicotinic acid were synthesized and evaluated for their inhibitory activity against COX-1 and COX-2, showing high potency and selectivity towards COX-2 inhibition. The most potent compound exhibited strong in vivo anti-inflammatory activity and potential influence on inflammatory cytokines, suggesting promising anti-inflammatory properties. Molecular docking studies further supported their remarkable COX-2 inhibitory activity compared to celecoxib.

Summary: Two series of twenty benzothiazole amide derivatives were synthesized and screened for their anti-inflammatory activity and gastric ulcerogenicity. Compounds 3h and 6a showed the most potent anti-inflammatory activity with high gastrointestinal tolerability. In vitro selectivity testing demonstrated that compound 6a had the best selectivity index among the investigated compounds.
