Stem cell-based therapies are considered to be a promising treatment method for several clinical conditions such as Alzheimer's disease, Parkinson's disease, spinal cord injury, and many others. However, the ideal stem cell type for stem cell-based therapy remains to be elucidated. Stem cells are present in a variety of tissues in the embryonic and adult human body. Both embryonic and adult stem cells have their advantages and disadvantages concerning the isolation method, ethical issues, or differentiation potential. The most described adult stem cell population is the mesenchymal stem cells due to their multi-lineage (trans)differentiation potential, high proliferative capacity, and promising therapeutic values. Recently, five different cell populations with mesenchymal stem cell characteristics were identified in dental tissues: dental pulp stem cells, stem cells from human exfoliated deciduous teeth, periodontal ligament stem cells, dental follicle precursor cells, and stem cells from apical papilla. Each dental stem cell population possesses specific characteristics and advantages which will be summarized in this review. Furthermore, the neural characteristics of dental pulp stem cells and their potential role in (peripheral) neural regeneration will be discussed.

Cumulative exposure and lifetime cancer risk from diagnostic radiation in patients undergoing orthognathic surgery: a cross-sectional analysis

J. Ver Berne, C. Politis, E. Shaheen, R. Jacobs

Summary: In this study, the cumulative effective dose and cancer risk for orthognathic surgery patients were calculated. The results showed that the cumulative effective dose was relatively low and the lifetime attributable risk of cancer was small compared to the baseline risk. These results provide a framework for interpreting radiation doses and cancer risks in orthognathic surgery patients.

Effect of foraminal enlargement on microcrack formation and apical transportation: a nano-CT assessment

Jader Camilo Pinto, Karla de Faria-Vasconcelos, Andre Ferreira Leite, Mariano Simon Pedano, Juliane Guerreiro-Tanomaru, Reinhilde Jacobs, Mario Tanomaru-Filho

Summary: The aim of this study was to evaluate the effects of foraminal enlargement on microcrack formation and apical transportation in root canals with apical curvature. Maxillary lateral incisors with apical curvature were divided into two groups, with one group undergoing foraminal enlargement. The results showed that foraminal enlargement led to the formation of new microcracks and increased apical transportation. Compared to root canal preparation up to 1 mm short of the apical foramen, foraminal enlargement using a heat-treated reciprocating file size 40 resulted in higher microcrack formation and apical transportation.

Summary: The aim of this study was to evaluate the image quality of CBCT exposure protocols and devices in detecting cracks and fine endodontic structures with metallic artifacts. Ten CBCT devices were used to scan an anthropomorphic phantom containing teeth with various structures. Different conditions were created by placing metallic objects next to the teeth of interest. The results showed that only specific CBCT devices and protocols were able to visualize cracks and fine structures effectively, while the presence of metallic artifacts significantly affected the image quality.

Is use of CBCT without proper training justified in paediatric dental traumatology? An exploratory study

Gertrude Van Gorp, Arno Maes, Marjan Lambrechts, Reinhilde Jacobs, Dominique Declerck

Summary: This study found that paediatric dentists had poor diagnostic accuracy when using CBCT without specific training, with higher sensitivity observed when using 2D images. Sensitivity was high for diagnosing apical pathology and root fractures, but low for inflammatory root resorption, root cracks, and subluxations. Additional training in radiographic diagnosis is necessary for paediatric dentists to improve diagnostic performance, especially when using CBCT.

Organoids from mouse molar and incisor as new tools to study tooth-specific biology and development

Florian Hermans, Lara Hemeryck, Celine Bueds, Marc Torres Pereiro, Steffie Hasevoets, Hiroto Kobayashi, Diether Lambrechts, Ivo Lambrichts, Annelies Bronckaers, Hugo Vankelecom

Summary: Organoid models have been successfully established to study tissue biology and development in a dish. In this study, tooth organoids (TOs) were developed from mouse molar and incisor, which express dental epithelium stem cell markers and recapitulate key properties of dental epithelium. These TOs can differentiate into ameloblast-resembling cells and co-differentiate into different cell types in the presence of dental mesenchymal stem cells. Furthermore, the TOs can also survive and differentiate in vivo. These organoid models provide new tools to study mouse tooth biology and development and may have implications for future human tooth repair and replacement.
