Conformation of the Solute-Binding Protein AdcAII Influences Zinc Uptake in Streptococcus pneumoniae

Summary: The pathogen Streptococcus pneumoniae acquires essential zinc ions from the host through the proteins AdcA and AdcAII, which is crucial for infection. AdcAII plays a larger role during initial infection, and the Zn(II)-coordinating residues are found to be important for metal binding. Through structural and biochemical analyses, it was shown that specific conformational changes in AdcAII are necessary for effective metal uptake.

Summary: This study demonstrates that extracellular vesicles produced by different serotypes of Streptococcus pneumoniae can be internalized by macrophages through fusion with host cell membranes, and have the ability to induce an immune response and promote the production of proinflammatory cytokines, with higher levels observed in certain serotypes.

Summary: A multicentre, retrospective observational study in eight Latin American and Caribbean countries assessed the direct effect of pneumococcal conjugate vaccines (PCVs) on the distribution of pneumococcal serotypes causing invasive pneumococcal disease in children younger than 5 years. The study found a significant decrease in the percentage of vaccine serotypes in PCV countries after PCV introduction.

Summary: In organ samples from deceased infants in India and Pakistan evaluated with multiplex polymerase chain reaction, Acinetobacter baumannii was the most common pathogen identified. Group B streptococcus was rarely found. A. baumannii was rarely found in the placentas of these deceased neonates, suggesting nosocomial acquisition.

Summary: Streptococcus pneumoniae shows different physiological and virulence responses to different carbohydrate availabilities. Differences in carbohydrate availability between the nasopharynx and invasive disease sites directly influence the metabolic activity, growth rate, and physiological state of S. pneumoniae. This anatomical site-specific carbohydrate availability directly affects the virulence traits of S. pneumoniae and promotes relative fitness.

Summary: Influenza A virus infection increases susceptibility to secondary bacterial pneumonia, with GP96 on infected cell surfaces mediating interactions with Streptococcus pneumoniae, leading to increased bacterial adherence and translocation. Inhibition of GP96 enhances pneumococcal clearance and reduces lung pathology in infected mice, suggesting a potential therapeutic target for inhibiting bacterial colonization in an IAV-infected respiratory tract.

Summary: This paper describes promising second-generation beta-lactamase inhibitors, such as avibactam, vaborbactam, and boronic acids, as well as several BL-BLI combinations. While some combinations are in clinical use, most are still limited to clinical trials or preclinical studies, highlighting the ongoing need for the development of novel BLIs to combat multidrug-resistant bacteria. Second-generation BLIs show promise as an effective approach in addressing antibiotic resistance.
