Summary: This study identified dominant pathogenic variants in LMNB1 and LMNB2 as a genetic cause of primary microcephaly, revealing a new form of laminopathy. The unique nature of the lamin B-associated phenotype highlights different developmental requirements for lamin paralogs and suggests a novel mechanism for primary microcephaly that warrants further investigation.

Small Molecule Inhibitors of Interferon-Induced JAK-STAT Signalling

Summary: Interferons (IFNs), important cytokines, play roles in triggering downstream biochemical events upon binding to cell surface receptors. Dysregulated IFN signaling has been linked to cancer progression and autoimmune diseases. Researchers have identified a small molecule that can block the activation of IFN signaling and further developing a more potent analogue that shows potential therapeutic effects in reducing bleeding in a model of hemorrhagic colitis. This first-in-class small molecule also inhibits other types of IFN-induced signaling. This research provides a foundation for the development of pan-IFN inhibitory drugs.

Summary: This study demonstrates the crucial role of Rab44 in regulating secretory granule transport during degranulation in mast cells. Deficiency in Rab44 impairs granule translocation and degranulation, leading to reduced sensitivity to IgE-mediated allergic reactions. Rab44 can be considered a potential target for modulating MC degranulation and inhibiting allergic reactions.

Summary: This study identified several ACAN gene variants associated with skeletal disorders, which affect the G3 domain of aggrecan proteoglycan. These variants lead to abnormal cartilage development and the occurrence of osteochondritis dissecans. Functional studies showed that the variant proteins were unable to be secreted to the same level as wild-type proteins and had decreased binding to cartilage extracellular matrix ligands.

Traboulsi syndrome caused by mutations in ASPH: An autosomal recessive disorder with overlapping features of Marfan syndrome

Summary: Traboulsi syndrome, a rare genetic disorder characterized by ocular and facial abnormalities, is associated with mutations in the ASPH gene. In addition to the characteristic eye and face features, patients may also experience aortic root dilation and inguinal hernias. This syndrome shares clinical features with Marfan syndrome and echocardiography surveillance is recommended for Traboulsi syndrome patients.

Summary: This study elucidates the genetic spectrum of Uncombable Hair Syndrome (UHS) and establishes a molecular genetic analysis of the largest worldwide collective of affected individuals. The findings suggest that pathogenic variants in PADI3, TCHH, and TGM3 could introduce molecular diagnostics for UHS, providing a new avenue for clinicians and affected individuals.
