Summary: In this study, polymerization mixture was optimized to synthesize poly(acrylamide-co-N,N'-methylenebisacrylamide) monolithic stationary phases for HILIC of intact proteins. The monolithic structure exhibited a permeable and polar stationary phase suitable for HILIC. The monolith showed superior separation performance and lower ion interaction compared to silica-based particulate amide phase, enabling successful hyphenation with high-resolution mass spectrometry in HILIC of intact proteins.

Summary: A simple, fast, and sensitive tandem mass spectrometry method was established to detect the concentration of regadenoson in human plasma, which was used to study the pharmacokinetic characteristics of regadenoson in healthy Chinese subjects. The method showed linear calibration curve, low precision, high accuracy, and no matrix effect for regadenoson or regadenoson-d3.

Summary: In this study, three recently developed HILIC columns were characterized in detail to compare their peptide retention mechanisms, revealing a multimodal retention mechanism in systems with these stationary phases. By balancing the properties of analytes and stationary phases, satisfactory retention and peak shape of peptides can be achieved. Buffer concentration and pH optimization was confirmed to be an effective tool for manipulating retention and peak symmetry, enhancing the potential applications of these columns in the analysis of diverse compounds.

Summary: In this study, GSL subclasses were isolated from human plasma using an optimized ethanol-water solvent system, followed by purification and analysis using HILIC-ESI-LIT-MS/MS. The results provide detailed profiling and characterization of GSL and phospholipids in human plasma, with the identification of 154 GSL species and 77 phospholipids. The developed method has potential applications in clinical and biological research of GSL in human blood and other biological samples.

Summary: This study systematically investigates the effects of different lipid class separation methods and mass spectrometers on quantitative lipidomic data, finding slight variations in molar concentrations even within the same laboratory using different platforms. Normalization to a reference sample can significantly reduce these differences, leading to highly homogenous molar concentrations of individual lipid species. Summary: Ovalbumin is a significant protein source, but the safety concerns of protein allergy and contamination have increased. Hence, it is necessary to detect proteins efficiently and rapidly. Mass spectrometry is a powerful tool used in protein detection. In this study, COF@S-S@GC was prepared and showed sensitive detection, good selectivity, high adsorption capacity, stability, reproducibility, linearity, LOQ level, and recovery ratio for ovalbumin. Furthermore, COF@S-S@GC also demonstrated satisfactory purification ability for protein enrichment from complex samples, indicating its great potential in protein analysis.

Summary: Crystallization-based separation techniques are commonly used in the polyolefin industry for CCD measurement. A new technique, iCCD, was developed using different separation packing materials to minimize cocrystallization and improve sample throughput rate. The iCCD method demonstrated equivalent cocrystallization degree to the industry standard method but with significantly improved sample throughput rate and measurement accuracy.

Summary: Nickel (Ni) is a trace heavy metal with documented allergy and carcinogenic effects in humans. Understanding the coordination mechanisms and labile complex species of Ni(II) is crucial for understanding its biological effects. Histidine (His) is an essential amino acid that contributes to protein structure and coordination of Ni(II) ions. This study evaluated different chromatographic columns for the separation of Ni(II)-Histidine species, and the Zic-cHILIC column showed high efficiency and selectivity. The identities of the Ni(II)-Histidine species were confirmed using HILIC-ESI-MS.

Summary: In order to address the challenges in analyzing certain biomolecules using liquid chromatography, researchers have developed a new hydrophilically modified hybrid surface (h-HST) for size exclusion chromatography and anion exchange separations. By incorporating additional oxygen and carbon atoms onto an ethylene bridge hybrid siloxane polymer, this surface reduces non-specific adsorption and undesired secondary interactions, resulting in improved separations of proteins and nucleic acids.

Summary: A sensitive and simple LC-MS/MS method was developed for the determination of orelabrutinib in human plasma and cerebrospinal fluid. The method involved simple protein precipitation, efficient separations, and optimized mass spectrometric conditions to increase selectivity and sensitivity. The method was validated for accuracy, precision, selectivity, linearity, recovery, matrix effect, stability, and limits of quantification. It exhibited a lower limit of quantification of 0.50 ng/mL, with intraday and interday precision RSD both less than 15%, and a recovery rate of 85.7%-92.9%. The method was successfully applied for quantitation in clinical patients treated with orelabrutinib.
