Summary: In this study, a generic HILIC-MS/MS method was developed for in-depth characterization of nucleic acid therapeutic modalities. High-resolution mass spectrometry was used to determine the deconvoluted masses of oligonucleotide and siRNA standards and their impurities. Optimized higher-energy C-trap dissociation was used to improve the sequence coverage of DNA and RNA oligonucleotides, achieving higher on-column sensitivity for phosphorothioate oligonucleotide analysis.

Summary: Aconitum, a highly toxic plant, contains alkaloids such as aconitine, hypaconitine, jesaconitine, and mesaconitine, which can cause fatal intoxication when ingested. It is crucial to analyze these alkaloids and their metabolites in the blood. This study employed QuEChERS extraction and liquid chromatography-tandem mass spectrometry to evaluate aconitines and their metabolites in postmortem blood samples. The findings suggest that aconitine and jesaconitine accumulate in the right atrium blood after death, and the right atrium specimen is suitable for measuring aconitine compounds in fatal intoxication cases.

Summary: This study aimed to determine the presence of promethazine (PMZ) and its metabolites, promethazine sulfoxide (PMZSO) and monodesmethyl-promethazine (Nor(1)PMZ), in different swine tissues. A sample preparation method and an analysis using high-performance liquid chromatography-tandem mass spectrometry (LC-MS/MS) were established and validated. The method showed good sensitivity, with limits of detection (LOD) and limits of quantification (LOQ) of 0.05 µg/kg and 0.1 µg/kg, respectively. The study also demonstrated good linearity and average recoveries for the target compounds, making it suitable for monitoring veterinary drug residues in animal-derived foods.

Summary: An efficient analytical method for the simultaneous determination of penthiopyrad enantiomers and its metabolite in different matrices was developed in this study, utilizing chiral stationary phase and ultra-high performance liquid chromatography-tandem mass spectrometry. The absolute configuration of penthiopyrad enantiomers was confirmed, and optimal conditions for chiral separation were determined through evaluation of different polysaccharide-based chiral stationary phases. The method showed good linearity, sensitivity, accuracy, and convenience for further research on stereoselective degradation, residual monitoring, and risk assessment.

Summary: Currently, MS/MS is widely used in metabolomics for the identification of metabolites. However, the structural identification of isomers remains challenging due to the similarity of their MS/MS spectra. Differentiation and identification of isomers using MS/MS can be facilitated by various techniques, including chemical derivatization, ion mobility spectrometry, ion dissociation techniques, and energy-resolved MS. This tutorial provides a comprehensive summary of these cutting-edge techniques and emphasizes the importance of isomeric identification in metabolomics for analytical chemists. Summary: Understanding the metabolic abnormalities of tumors is crucial for diagnosis, prognosis, and treatment. However, analyzing aberrant metabolites in tumors using common techniques like LC-MS and GC-MS face challenges due to their polarity, isomerism, or low ionization efficiency. Chemical derivatization of metabolites provides an effective solution to overcome these challenges. This review focuses on the difficulties encountered, the principles behind chemical derivatization methods, and the advancements in analyzing tumor metabolites using derivatization-based chromatography.
