Suppression of Liver Tumor Growth and Metastasis by Adiponectin in Nude Mice through Inhibition of Tumor Angiogenesis and Downregulation of Rho Kinase/IFN-Inducible Protein 10/Matrix Metalloproteinase 9 Signaling

Purpose: We aimed to investigate the effects of adiponectin on liver cancer growth and metastasis and explore the underlying mechanisms. Experimental Design: An orthotopic liver tumor nude mice model with distant metastatic potential was applied. Either Ad-adiponectin (1 x 10(8); treatment group) or Ad-luciferase (control group) was injected via portal vein after tumor implantation. Tumor growth and metastasis were monitored by Xenogen In vivo Imaging System. Hepatic stellate cell activation by a-smooth muscle actin staining, microvessel density by CD34 staining, macrophage infiltration in tumor tissue, and cell signaling leading to invasion, migration [Rho kinase (ROCK), IFN-inducible protein 10 (IP10), and matrix metalloproteinase 9], and angiogenesis [vascular endothelial growth factor (VEGF) and angiopoietin 1] were also compared. Tumor-nontumor margin was examined under electron microscopy. Direct effects of adiponectin on liver cancer cells and endothelial cells were further investigated by a series of functional studies. Results: Tumor growth was significantly inhibited by adiponectin treatment, accompanied by a lower incidence of lung metastasis. Hepatic stellate cell activation and macrophage infiltration in the liver tumors were suppressed by adiponectin treatment, along with decreased microvessel density. The treatment group had less Ki-67-positive tumor cells and downregulated protein expression of ROCK1, proline-rich tyrosine kinase 2, and VEGF. Tumor vascular endothelial cell damage was found in the treatment group under electron microscopy. In vitro functional study showed that adiponectin not only downregulated the ROCK/IP10/VEGF signaling pathway but also inhibited the formation of lamellipodia, which contribute to cell migration. Conclusion: Adiponectin treatment significantly inhibited liver tumor growth and metastasis by suppression of tumor angiogenesis and downregulation of the ROCK/IP10/matrix metalloproteinase 9 pathway. Summary: A growing body of evidence suggests that the gut microbiota plays a role in the pathogenesis of nonalcoholic fatty liver disease (NAFLD). This study found that machine learning models integrating microbial signatures were able to accurately classify individuals based on their NAFLD status and liver fat accumulation, outperforming other clinical models. These findings raise the possibility of using gut microbiota for early clinical warning of NAFLD development.

Summary: We aimed to investigate the relationship of B2M, cystatin C, and LCN-2 with stroke risk in a general Chinese population. Elevated levels of serum B2M, cystatin C, and LCN-2 were significantly associated with stroke risk. These biomarkers may serve as useful tools for clinicians to assess stroke risk.

The Landscape of Aberrant Alternative Splicing Events in Steatotic Liver Graft Post Transplantation via Transcriptome-Wide Analysis

Summary: The application of steatotic liver graft has been increased due to donor shortage and prevalence of non-alcoholic fatty liver disease. However, steatotic livers are vulnerable to inflammatory injury, which may lead to cancer recurrence. This study identified differential alternative splicing events (ASEs) in steatotic liver grafts and their association with tumor recurrence. The differential ASEs were associated with HCC patients' survival and were enriched in metabolic pathways and immune cell distribution. The cancer hallmarks were also enriched in steatotic liver grafts and linked to differential ASEs. This work provides new insights into the relationship between steatotic liver graft injury and tumor recurrence, as well as potential markers for prognosis and therapy.

Circulating AFABP, FGF21, and PEDF Levels as Prognostic Biomarkers of Sight-threatening Diabetic Retinopathy

Summary: This study evaluated the associations of three diabetes-related biomarkers (AFABP, FGF21, and PEDF) with the development of sight-threatening diabetic retinopathy (STDR). The results showed that serum AFABP level was independently associated with incident STDR and its addition to a clinical model improved the prediction of STDR among participants without baseline retinopathy.

Summary: This study aimed to investigate the association between serum TSP2 level and kidney function decline in patients with type 2 diabetes. The results showed that serum TSP2 level was independently associated with incident eGFR decline and could predict a rapidly declining trajectory. This finding is of great importance for the renal health management of patients with diabetes.

Chronic SIRT1 supplementation in diabetic mice improves endothelial function by suppressing oxidative stress

Summary: This study aimed to investigate the therapeutic potential of chronic recombinant murine SIRT1 (rmSIRT1) supplementation to alleviate endothelial and vascular dysfunction in diabetic mice. The results showed that arterial SIRT1 levels were significantly reduced in diabetic conditions, while rmSIRT1 supplementation restored SIRT1 levels and improved endothelial function and vascular compliance. The study suggests that SIRT1 supplementation may represent a novel therapeutic strategy to prevent and treat diabetic vascular disease by enhancing eNOS activity and suppressing NOX-related oxidative stress.

Post-transplant inflammatory cytokine signature adds value for predicting tumor recurrence after liver transplantation for hepatocellular carcinoma

Summary: This study found that post-transplant cytokines CCL11, IFN alpha 2, and IL17A are significant predictors of tumor recurrence and survival after liver transplantation for hepatocellular carcinoma. A prediction model including these cytokines, the UCSF criteria, and pre-transplant AFP accurately predicted post-transplant tumor recurrence and guided refinements in surveillance and therapeutic strategies.

Summary: This study aimed to develop liquid biopsy assays for early HCC diagnosis and prognosis. A panel of 23 microRNAs (HCCseek-23 panel) was consolidated based on their reported functions in HCC development. Serum samples were collected from 103 early-stage HCC patients before and after hepatectomy. Quantitative PCR and machine learning random forest models were applied to develop diagnostic and prognostic models.
