Summary: A new humanized anti-IL-6 mAb HZ-0408b with potent binding and neutralizing activity to human IL-6 has been developed. It showed a high species specificity and lower cross-reactivity compared to other anti-IL-6 mAbs. HZ-0408b exhibited a stronger inhibitory effect on IL-6 signaling and effectively reduced joint swelling and plasma CRP levels in a monkey CIA model.

Anti-IL-6 receptor antibody could overcome tumor immunosuppression and inhibit tumor progression.

In vitro experiments showed that recombinant HMGB1 delayed hemoglobin clearance, allowing the toxic effects of hemoglobin, heme, and Fe2 + to persist. Additionally, humanized anti-HMGB1 mAb reduced body weight loss and improved behavioral performance after ICH.

Summary: In this study, a humanized and Fc-engineered anti-B7-H3 mAb 24F-Hu-mut2 was developed and its anti-tumor activity was systematically evaluated in vitro and in vivo. The results showed significant anti-ESCC activity of 24F-Hu-mut2. A computational docking model and ELISA results confirmed the binding of the mAb to specific domains of B7-H3. This study suggests that 24F-Hu-mut2 may be a promising antibody for treating ESCC and other B7-H3 overexpressing tumors.

Summary: This study investigates the potential of a combination therapy involving an anti-IL-8 antibody and an anti-PD-1 antibody for treating pancreatic ductal adenocarcinoma (PDAC). The results show that the combination treatment enhances the antitumor activity and activates the innate immune response in the tumor microenvironment. CD16+ myeloid cells, particularly granulocytes, are identified as the main target of IL-8. These findings provide a basis for further clinical testing of this combination therapy in PDAC treatment and highlight the role of peripherally derived myeloid cells in promoting the antitumor response of T cells.
