Summary: Colon-targeted trans-cinnamic acid (tCA) and its conjugates with glutamic acid (tCA-GA) and aspartic acid (tCA-AA) were designed and synthesized. The anti-colitic activity of colon-targeted tCA was evaluated using a rat colitis model. The conjugates decreased the distribution coefficient and cell permeability of tCA and produced more tCA in the cecum, particularly with tCA-GA. Oral administration of tCA-GA delivered a higher amount of tCA to the cecum and exhibited better anti-colitic effects compared to tCA and the current treatment, sulfasalazine (SSZ).

Summary: Drug tolerance and low efficiency of single treatment have negatively impacted the survival time of colorectal cancer patients. The combination of chloroquine and PD-1 siRNA shows promising results in inhibiting tumor growth, metastases, and inducing apoptosis, while enhancing the immune response. This combined approach represents a new avenue for clinical treatment of colorectal cancer.

Summary: The scale-up of a chiral bicyclic homopiperazine of pharmaceutical interest was improved using continuous-flow chemistry, with enhancements in outcome and safety profile of a key batch ring-expansion step. The selectivity of nitrogen insertion for the ring expansion was enhanced via an alternative photochemical oxaziridine rearrangement under mild conditions, leading to the first photochemical scale-up of a homopiperazine in a simple and efficient flow reactor.

Summary: In the past decade, significant progress has been made in new cancer treatments for melanoma, particularly in on-treatment survival. However, overall survival rates have improved relatively modestly. Melanoma exhibits heterogeneity and transcriptional plasticity, enabling it to adapt and escape even advanced treatments. Although our understanding of melanoma biology and genetics has advanced, debates continue regarding the cell of origin for melanoma. Animal models and single-cell sequencing approaches offer new avenues of research in addressing this question.
