Safety and immunogenicity of co-administered MF59-adjuvanted 2009 pandemic and plain 2009-10 seasonal influenza vaccines in rheumatoid arthritis patients on biologicals

Rheumatoid arthritis (RA) patients under immunosuppressive therapy are particularly susceptible to infections, mainly of the respiratory tract, thus vaccination may represent a strategy to reduce their incidence in this vulnerable population. In the 2009-10 influenza season, the safety and immunogenicity of co-administered non-adjuvanted seasonal and MF59-adjuvanted pandemic influenza vaccines were evaluated in this study in 30 RA patients under therapy with anti-tumour necrosis factor (TNF)- agents or Abatacept and in 13 healthy controls (HC). Patients and HC underwent clinical and laboratory evaluation before (T0), 1 (T1) and 6 months (T2) after vaccinations. No severe adverse reactions, but a significant increase in total mild side effects in patients versusHC were observed. Both influenza vaccines fulfilled the three criteria of the Committee for Proprietary Medicinal Products (CPMP). Seroconversion rate for any viral strain in patients and HC was, respectively, 68 versus 45 for H1-A/Brisbane/59/07, 72 versus 81 for H3-A/Brisbane/10/07, 68 versus 54 for B/Brisbane/60/08 and 81 versus 54 for A/California/7/2009. A slight increase in activated interferon (IFN)--, TNF-- or interleukin (IL)-17A-secreting T cells at T1 compared to T0, followed by a reduction at T2 in both patients and HC, was registered. In conclusion, simultaneous administration of adjuvanted pandemic and non-adjuvanted seasonal influenza vaccines is safe and highly immunogenic. The largely overlapping results between patients and HC, in terms of antibody response and cytokine-producing T cells, may represent further evidence for vaccine safety and immunogenicity in RA patients on biologicals.

Melanoma Cell Resistance to Vemurafenib Modifies Inter-Cellular Communication Signals

Summary: The study focused on secretome profiling of melanoma cells sensitive or resistant to the BRAFi vemurafenib, finding that vemurafenib-resistant melanoma cells can influence dendritic cell maturation and produce higher levels of pro-inflammatory cytokines. Proteomic and cytokine/chemokine secretion analyses suggest a possible involvement of the proteasome pathway in BRAFi resistance, with BRAFi-resistant cells displaying a more aggressive phenotype and increased production of specific proteins. Plasma levels of CD147/basigin and MMP-2 were observed to increase in melanoma patients at the time of treatment failure, supporting the hypothesis that CD147/basigin might play a role in BRAFi resistance.

Summary: In this study, we analyzed the SARS-CoV-2 genomes from different regions in Italy. The results showed that the spread primarily occurred from the north to the center and south, consistent with epidemiological data. The estimated evolutionary rate was similar to previous studies, and the phylogenetic analysis suggested that the virus entered Italy in late January or early February 2020. Continuous molecular surveillance is necessary to track virus circulation and evolution.

Reduction of the risk of severe COVID-19 due to Omicron compared to Delta variant in Italy (November 2021-February 2022)

Summary: This study aimed to assess the impact of the Omicron variant on severe COVID-19 compared to the Delta variant in Italy. The study found that cases with Omicron had a reduced risk of severe COVID-19 compared to Delta. The protective effect was most significant in the 40-59 age group, while no protection was observed in the 12-39 age group. Vaccination was associated with a lower risk of severe COVID-19 in both variants.

Summary: The Istituto Superiore di Sanita (ISS) established a Working Group on Scientific Literature updates (WG SL) during the Sars-CoV-2 pandemic, screening a large number of pre-prints and peer reviewed papers to provide real-time knowledge and evidence for health-workers. They submitted daily summary reports of pre-print selection to the ISS President and published weekly open access papers of interest for health professionals, monitored through satisfaction questionnaires. Promoting health literacy with a cross-cutting approach has been proven to be an effective non-pharmacological intervention.
