Electrochemotherapy is effective in the treatment of rat bone metastases

Bone metastases impair general health status, quality of life and survival of patients. Electrochemotherapy (ECT), which combines electroporation (EP) and the administration of anticancer drugs, has been recently introduced into clinical practice for the local treatment of solid tumours. In the present study, the ability of EP with bleomycin (Bleo) to induce MRMT-1 rat breast cancer cell death was investigated in vitro. Then, an in vivo model for bone metastases was set up by the inoculation of MRMT-1 cells in rat proximal tibia. 7 days after tumour induction the animals were treated with Bleo, EP, Bleo followed by EP (ECT), or left untreated. ECT eliminated the tumour in 6 out of 8 (75 %) treated metastases. Radiological evaluation showed that the Honore score in ECT-treated animals was significantly lower when compared with the other groups (p < 0.0005) and not significantly different from healthy controls. Bone morphology in ECT-treated animals, evaluated by histological and microtomographical analyses, showed intact cortical and trabecular bone structure with new bone apposition. Histomorphometric evaluation showed that ECT-treated metastases had significantly higher bone volume, trabecular number, trabecular thickness and bone mineral density compared with those of untreated metastases (respectively p < 0.0005 for BV/TV, Tb.N and BMD; p < 0.05 for Tb.Th) or metastases treated with Bleo (p < 0.05 for BV/TV, Tb.N, p < 0.005 for BMD) or EP (p < 0.005 for BV/TV, Tb.N; p < 0.0005 for BMD). These findings suggest that early ECT treatment of bone metastases is minimally invasive, safe and effective, thus providing pre-clinical evidence for its use in the treatment of human bone metastases.

Bone Regeneration Guided by a Magnetized Scaffold in an Ovine Defect Model

Summary: The use of functionalized scaffolds incorporated with superparamagnetic nanoparticles and vascular endothelial growth factor (VEGF) injection into a magnetic scaffold resulted in increased bone regeneration in a large bone defect in sheep metatarsus. The magnetic field generated by implanted magnets allowed the capture of the injected nanoparticles, forming a VEGF gradient in the scaffold's porosity. Histomorphometric measurements showed improved bone growth and bone-to-implant contact, while atomic force microscopy (AFM) analysis revealed similar nanomechanical behavior of the regenerated tissue to the magnetic field distribution. This study demonstrates the potential of magnetic technologies combined with functionalized scaffolds to enhance bone formation and improve bone/scaffold interaction.

Bilayer Dense-Porous Li7La3Zr2O12 Membranes for High-Performance Li-Garnet Solid-State Batteries

Intermediate-Stage Sintered LLZO Scaffolds for Li-Garnet Solid-State Batteries

Complications after Posterior Lumbar Fusion for Degenerative Disc Disease: Sarcopenia and Osteopenia as Independent Risk Factors for Infection and Proximal Junctional Disease

Summary: Proximal junctional disease (PJD) and surgical site infection (SSI) are common complications after spine surgery. This study aimed to evaluate the influence of sarcopenia and osteopenia on these complications. Preoperative MRI was used to measure central sarcopenia and osteopenia. The analysis showed that comorbidity index and dural tear were independent risk factors for SSI, while age and diabetes were risk factors for PJD. However, there was no correlation between low M-score and PLVI and a higher complications rate.

Gender-Specific Differences in Human Vertebral Bone Marrow Clot

Summary: Recently, our group studied the use of vertebral bone marrow (vBMA) clot as a cell therapy for spinal fusion. We compared the properties of vBMA clots and derived vertebral mesenchymal stem cells (MSCs) from female and male patients who underwent spinal fusion procedures and received vBMA clot treatment. The results showed that vBMA clots and MSCs from males had higher expression of growth factors (GFs) and greater osteogenic and chondrogenic potential compared to females. The findings also revealed differences in the gene expression profiles of MSCs derived from vBMA clots between female and male donors.

A Systematic Review of the Retrograde Drilling Approach for Osteochondral Lesion of the Talus: Questioning Surgical Approaches, Outcome Evaluation and Gender-Related Differences

Proprioception and Mechanoreceptors in Osteoarthritis: A Systematic Literature Review

Summary: This review systematically analyzes the changes in proprioception and related mechanoreceptors in osteoarthritis (OA) patients. The studies found that OA patients may have impaired proprioceptive accuracy in the posterior cruciate ligament (PCL), possibly due to changes in mechanoreceptors. Significant proprioceptive deficits were also identified in patients with knee articular cartilage lesions. Further research is needed to clarify the role of proprioception and related mechanoreceptors in OA.
