Relationships between serum osteocalcin, leptin and the effect of weight loss by pharmacological treatment in healthy, nonsmoking Korean obese adults

Background: Recent studies have reported a relationship between osteocalcin (OC) levels and factors associated with energy metabolism and insulin resistance. As any detailed understanding of OC mechanisms still remains elusive, this study aimed at revealing a correlation between serum DC levels and obesity in healthy, nonsmoking, Korean obese adults who had undergone weight loss through pharmacological treatment. Methods: 119 healthy, nonsmoking, Korean obese adults were investigated at 3 months following weight loss through pharmacological treatment. Serum OC, leptin, HOMA score, ghrelin, visceral fat mass, total body fat, and BMI were measured. Results: Increase in serum OC was significantly associated with decreases in: BMI (and weight change %) (r=-0.209, p=0.023), visceral fat mass (r=-0.189, p=0.049), HOMA (r=-0.203 p=0.027), and leptin (r=-0.253 p=0.006), but not with changes in adiponectin (r=+0.029, p=NS), and Ghrelin (r=+0.019, p=NS). Decrease in leptin (beta=-0.280, p=0.002) was significantly associated with an increase in serum OC, after pharmacological weight loss treatment was adjusted for age, sex, drug type, and BMI (or visceral fat mass). Conclusions: Serum OC was significantly increased at 3 months after pharmacological weight loss. We further found that leptin levels were associated with changes in serum OC. These findings suggest a relationship between bone and adipose tissue. (C) 2013 Elsevier B.V. All rights reserved.

Summary: Through computational secretome analyses, we identified a blood biomarker AKR1B10 that reflects the progression of nonalcoholic fatty liver disease (NAFLD). Plasma AKR1B10 has a good diagnostic accuracy for nonalcoholic steatohepatitis (NASH) and fibrosis, and it can be further improved when combined with C3. Plasma AKR1B10 is also a specific biomarker for NAFLD progression.

Pharmacotherapy in obesity: the current state and the near future

Summary: The prevalence of obesity in Korea is rapidly increasing, and more effective treatment tools are needed. Currently approved drugs for obesity treatment include appetite suppressants, dietary fat absorption inhibitors, GLP-1 receptor agonists, and fixed-dose combination drugs. However, these drugs have limited weight reduction effects. Newly developed agents such as semaglutide and tirzepatide show promise in being more effective, but may cause gastrointestinal adverse events.

Summary: This study evaluated the benefits and harms of carbohydrate-restricted diets and intermittent fasting (IF) and provided recommendations based on scientific evidence. Moderate-low carbohydrate or low carbohydrate diets were suggested for weight reduction in adults with overweight or obesity. In adults with type 2 diabetes, moderate-low carbohydrate diets were recommended for improving glycemic control and reducing body weight. Very-low carbohydrate diets and IF were not recommended for patients with diabetes. No specific recommendations were given for very-low carbohydrate diets and IF in adults with overweight or obesity and carbohydrate-restricted diets and IF in patients with hypertension.

Trajectories of Handgrip Strength and Their Associations with Mortality among Older Adults in Korea: Analysis of the Korean Longitudinal Study of Aging

Summary: The study showed that decreased handgrip strength was associated with increased all-cause mortality among older adults. Different trajectory groups of handgrip strength were identified for men and women. Genetics of congenital heart disease

Both genetic and non-genetic factors contribute to the co-occurrence of CHD and NDD. Further research should focus on identifying common molecular mechanisms underlying this co-occurrence and promoting the research and treatment of developmental disorders related to the cardiovascular and central nervous systems.

Flow cytometry in the detection of circulating tumor cells in neoplastic effusions

Accurate identification of HLA-B*15:02 allele by two-dimensional polymerase chain reaction

Summary: This study successfully established a 2D-PCR method for identifying HLA-B*15:02 through a two-tube reaction. This method can distinguish HLA-B*15:02 from 16 highly homologous HLA-B*15 alleles. Among 1830 samples from the clinical general population, 3 HLA-B*15:02 homozygotes and 84 HLA-B*15:02 heterozygotes were detected.
