The aim of this study was to investigate apolipoprotein (apo) A-l, apo B, lipoprotein (Lp) (a), HDL-cholesterol (C), LDL-C, triglycerides (TG) and total cholesterol (TC) values in the serum and synovial fluid (SF) of untreated rheumatoid arthritis (RA), psoriatic arthritis (PsA), and osteoarthritis (OA) patients. Methods: Paired SF and serum samples were collected simultaneously from 14 patients with RA, 14 with PsA, and 16 with OA and tested for apo A-l, apo B, HDL-C, LDL-C, Lp(a), TC and TG. Serum C reactive protein (CRP) and amyloid A (SAA) levels were also determined. Results: The inflammatory arthritis patients had higher SF lipid levels with the exception of HDL Reflecting increased synovial permeability, the lipid SF/serum ratio was always higher in RA and PsA with respect to OA patients. The positive correlation between serum and SF apo A-l, apo B. HDL-C, TG, and Lp(a) levels confirmed that there is lipoprotein diffusion into the SF. RA and PsA patients had lower concentrations of all serum lipids except for Lp(a) with respect to OA patients. The levels in the RA patients were similar to those in healthy matched controls, while the PsA patients had significantly lower apo A-l and HDL levels and higher apo B and LDL values. Conclusions: Lipid diffusion into the joint cavity, which largely depends on the degree of inflammation, may contribute to modulating local inflammatory processes. (C) 2011 Elsevier B.V. Bactericidal/Permeability-Increasing Protein Downregulates the Inflammatory Response in In Vivo Models of Arthritis

Anna Scanu, Roberto Luisetto, Francesca Oliviero, Francesca Galuppini, Vanni Lazzarin, Gianmaria Pennelli, Stefano Masiero, Leonardo Punzi

Summary: The study showed that BPI alone or in combination with HA can improve inflammation in animal models of CIA and crystal-induced inflammation. The combined use of BPI and HA resulted in greater improvement in both models compared to the administration of the substances alone.

Seasonal variation and disease distribution of cytophagocytic mononuclear cells in synovial fluid: A medical record study

Oliviero Francesca, Galozzi Paola, Padoan Andrea, Lorenzin Mariagrazia, Scanu Anna, Ramonda Roberta

Summary: This study investigated the frequency and disease distribution of cytophagocytic mononuclear (CPM) cells in synovial fluid of patients with rheumatic diseases over a 12-year period. The results showed a strong association between CPM cells and seronegative spondyloarthritis (SpA), with a seasonal variation in their incidence, peaking in spring and autumn. This suggests that the presence of CPM cells in synovial fluid may offer diagnostic insights into SpA.

Leucocyte Abnormalities in Synovial Fluid of Degenerative and Inflammatory Arthropathies

Summary: Genomic damage is associated with autoimmune processes, chronic inflammation, and apoptosis. The T cell compartment may exhibit overall genomic instability in certain rheumatological diseases. However, there is limited knowledge regarding leukocyte abnormalities in synovial fluid (SF) and their connection to inflammation. This study aimed to investigate cellular phenotypes in SF from patients with various inflammatory arthropathies. The results showed that inflammatory SF is associated with genomic instability and abnormal cell subsets.

Summary: This study aimed to evaluate the association between fetuin-A levels and radiographic sacroiliitis and syndesmophytes in early axSpA patients, and to identify potential predictors of radiographic damage. Results showed that fetuin-A levels were negatively associated with sacroiliac joint damage and may serve as a biomarker for predicting early structural damage.

Short-Term TERT Inhibition Impairs Cellular Proliferation via a Telomere Length-Independent Mechanism and Can Be Exploited as a Potential Anticancer Approach

Aamir Amin, Marzia Morello, Maria Raffaella Petrara, Beatrice Rizzo, Francesco Argenton, Anita De Rossi, Silvia Giunco

Summary: Blocking telomerase (TERT) to erode telomeres and inhibit cancer cell proliferation seems impractical. However, evidence suggests that TERT has telomere-length independent functions that promote tumor growth. Understanding the mechanisms behind these effects is crucial in establishing TERT as a therapeutic target. By inhibiting TERT in B-cell lymphoproliferative disorders and malignancies, we found that it impairs the NF-kappa B p65 pathway, resulting in decreased MYC expression and cell cycle arrest. In vivo results in zebrafish models support these findings, indicating the potential of combining TERT inhibition with chemotherapy for effective cancer treatment.

Summary: This review explores the potential benefits of balneotherapy (BT) as a complementary intervention for managing hand osteoarthritis (HOA), based on recent randomized controlled trials (RCTs). The findings indicate that BT, particularly when using specific methods such as mud packs and mineral-rich baths, shows promise in reducing pain and improving functionality and quality of life, especially in the initial stages of the condition. Further research and larger-scale trials are needed to validate these preliminary findings.

Summary: The cardiovascular system and the central nervous system exhibit a coordinated developmental process during embryonic development. Congenital heart disease (CHD) is the most common congenital disorder, and neurodevelopmental disorders (NDD) are common complications in CHD patients. Both genetic and non-genetic factors contribute to the co-occurrence of CHD and NDD. Further research should focus on identifying common molecular mechanisms underlying this co-occurrence and promoting the research and treatment of developmental disorders related to the cardiovascular and central nervous systems.
