Myeloid Cell-Specific ABCA1 Deletion Protects Mice From Bacterial Infection

Rationale: ATP-binding cassette transporter A1 (ABCA1) plays a critical role in eliminating excess free cholesterol from tissues by effluxing cellular free cholesterol and phospholipids to lipid-poor apolipoprotein AI. Macrophage ABCA1 also dampens proinflammatory myeloid differentiation primary-response protein 88-dependent toll-like receptor signaling by reducing cellular membrane free cholesterol and lipid raft content, indicating a role of ABCA1 in innate immunity. However, whether ABCA1 expression has a role in regulating macrophage function in vivo is unknown. Objective: We investigated whether macrophage ABCA1 expression impacts host defense function, including microbial killing and chemotaxis. Methods and Results: Myeloid cell-specific ABCA1 knockout (MSKO) vs wild-type mice were infected with Listeria monocytogenes (Lm) for 36 hours or 72 hours before euthanasia. Lm-induced monocytosis was similar for wild-type and MSKO mice; however, MSKO mice were more resistant to Lm infection, with significantly less body weight loss, less Lm burden in liver and spleen, and less hepatic damage 3 days postinfection. In addition, Lminfected MSKO mouse livers had: (1) greater monocyte chemoattractant protein-1 and macrophage inflammatory protein-2 expression; (2) more monocyte/macrophage infiltration; (3) less neutral lipid accumulation; and (4) diminished expression of lipogenic genes. MSKO macrophages showed enhanced chemotaxis toward chemokines in vitro and increased migration from peritoneum in response to lipopolysaccharide in vivo. Lm infection of wildtype macrophages markedly reduced expression of ABCA1 protein, as well as other cholesterol export proteins (such as ATP-binding cassette transporter G1 and apolipoprotein E). Conclusions: Myeloid-specific ABCA1 deletion favors host response to and clearance of Lm. Macrophage Lm infection reduces expression of cholesterol export proteins, suggesting that diminished cholesterol efflux enhances innate immune function of macrophages. Recent Advances on Small-Molecule Bromodomain-Containing Histone Acetyltransferase Inhibitors

Summary: In recent years, extensive research has been carried out on the molecular mechanisms and inhibitors targeting bromodomains (BRDs) and extra-terminal (BET) family proteins. Non-BET BRDs are gaining attention as a research hot spot. BRDs are abundant in histone acetyltransferase (HAT)-associated activating transcription factors, and BRD-containing HATs have been associated with cancer, inflammation, and viral replication. The development of BRD-containing HATs as chemical probes is valuable for understanding the specific biological roles of BRDs in diseases and drug discovery. Different types of BRD-containing HATs, including CBP/P300, PCAF/GCN5, and TAF1, are discussed in terms of their structures, functions, and small-molecule inhibitors. Furthermore, the progress in BRD inhibitors/chemical probes and proteolysis targeting chimeras regarding drug design, biological activity, and disease application is summarized. These findings provide insights into the development of BRD inhibitors as potential drug candidates for various diseases.

Unsupervised cross-domain functional MRI adaptation for automated major depressive disorder identification

Summary: This study proposes a novel discrepancy-based unsupervised cross-domain fMRI adaptation framework (UFA-Net) for automated identification of major depressive disorder (MDD) using resting-state functional magnetic resonance imaging (rs-fMRI) data. The UFA-Net models the spatio-temporal fMRI patterns of labeled source and unlabeled target samples and aligns features between two domains using attention-guided graph convolution module and maximum mean discrepancy constrained module. The proposed method outperforms several state-of-the-art methods and can facilitate fMRI-based analysis of MDD in clinical practice.

Association between polymorphisms of the GSDMB gene and allergic rhinitis risk in the Chinese population: a case-control study

Summary: This study found that SNPs in GSDMB gene are associated with susceptibility to allergic rhinitis (AR), with rs4795400 showing a protective effect in overall, males, people with BMI ≤ 24, and those living in wind-blown sand area. rs2305479 is associated with reduced risk of AR in males, while rs12450091 is a risk factor for AR in people living in the loess hilly region. The findings suggest that GSDMB polymorphisms are correlated with AR susceptibility.

DomainATM: Domain adaptation toolbox for medical data analysis

Summary: Domain adaptation is an important technique in machine learning-based medical data analysis to reduce distribution differences between different datasets. The Domain Adaptation Toolbox for Medical data analysis (DomainATM) is an open-source software package implemented in MATLAB, offering a collection of popular data adaptation algorithms for medical image analysis. It provides researchers with the capability to perform fast feature-level and image-level adaptation, visualization, and performance evaluation of adaptation methods. Users can also develop and test their own adaptation methods through scripting, enhancing the utility and extensibility of DomainATM. The software, source code, and manual are available online.

Epitaxial Growth of Hollow Gold Nanotetrakaidecahedrons on Hollow Gold Nanocubes: A Method for Modifying the Morphology of Hollow Nanoparticles

Summary: The morphology of hollow noble metal nanoparticles is crucial for their unique properties and applications. In this study, a galvanic replacement reaction was used to synthesize hollow gold nanocubes, and ascorbic acid and CTAB were introduced to control the shape of the resulting hollow gold nanotetrakaidecahedrons. CTAB selectively adsorbed on specific facets, inhibiting their growth and allowing the {111} facet to grow fastest. Optical absorption spectra further confirmed the formation and growth mechanism. The study demonstrates the ability to fabricate hollow nanoparticles with desired shapes by adjusting experimental conditions.

The function of long non-coding RNA in non-alcoholic fatty liver disease

Detecting type 2 diabetes mellitus cognitive impairment using whole-brain functional connectivity

Summary: Research shows that the functional connectivity in patients with T2DM is associated with mild cognitive impairment, and rs-fMRI can be used for early diagnosis. By analyzing the rs-fMRI data of T2DM-MCI patients, T2DM-NCI patients, and normal controls, it was found that the thalamus, angular, caudate nucleus, and paracentral lobule had the greatest contribution to the classification outcome. These findings provide valuable knowledge for classification and prediction of T2DM-related cognitive impairment, aid in early clinical diagnosis, and lay the foundation for future studies.

Accurate module induced brain network construction for mild cognitive impairment identification with functional MRI

Summary: In this paper, an accurate module induced Pearson correlation (AM-PC) model is proposed to estimate functional brain networks (FBNs) with a clear modular structure by incorporating sparse and low-rank constraints on the Laplacian matrix of the network. Experimental results show that the proposed method achieves better classification performance for classifying subjects with mild cognitive impairment (MCI) from healthy controls compared to previous methods.

Summary: Chemical kinetic studies of hydrogen atom abstraction reactions by hydroperoxyl radical from six alkyl cyclohexanes are conducted through high-level ab initio calculations. Rate constants, branching ratios, and thermodynamic properties are obtained, and their effects on ignition delay time predictions and species concentrations are investigated. The investigated reactions promote ignition delay times and improve the prediction of cyclic olefin species formation.

Dual manipulation of light and shape based on nanoparticle-induced shape memory composites

The membrane is composed of thermo-irritated shape memory polymers and doped nonclose-packing nanoparticles (NPs). The programmable reconfiguration and transparency-changing of this smart film highlight its great potential in various applications.
