Constitutive repression of interleukin-1α in endothelial cells

Activation of complement stimulates inflammation and provides an initial vigorous defense against infection. Insertion of the membrane attack complex in cell membranes of vascular endothelial cells induces changes in cell differentiation that promote coagulation, thrombosis, inflammation, and immunity. These changes are mediated by production of interleukin (IL)-1 alpha by endothelial cells, which acts locally on endothelial cells to contain infection and promote healing of the affected site. In healthy tissues, however, promoting coagulation and inflammation would be dysphysiologic. Accordingly, endothelial cell activation by the membrane attack complex depends on both transcriptional regulation of IL-1 alpha and availability of that cytokine to broadly modify endothelial cell physiology. Here, we report that the IL-1 alpha gene contains a suppressor sequence that cooperates with histone modification to regulate production of IL-1 alpha by endothelial cells. The suppressor sequence binds C/EBP (CCAAT enhancer-binding protein) family DNA-binding proteins isolated from the nucleus of quiescent endothelial cells. These results suggest constitutive suppression of IL-1 alpha maintains quiescence of endothelium and that terminal complement complexes remove that suppression, allowing IL-1 alpha transcription and, ultimately, activation of endothelium to proceed.

Summary: Neutralizing antibodies against SARS-CoV-2 are considered to be the most immediate and effective treatment for severely afflicted individuals. Broadly neutralizing antibodies are especially sought due to the potential diversification of the coronavirus through mutations. A study reported a novel approach to rapidly generate potent broadly neutralizing human anti-SARS-CoV-2 antibodies, isolating spike protein-specific memory B cells from convalescent individuals and expressing them as human monoclonal antibodies. All 43 generated human monoclonal antibodies neutralized SARS-CoV-2, with some showing high potency against spike-pseudotyped virus and variants, suggesting potential resistance to immunity evoked by infection or vaccines developed based on the Wuhan Hu-1 strain.

Summary: Research has shown successful engraftment of immature human hepatocytes in immunodeficient swine, with evidence of adaptive immune response in fetuses. However, rejection of xenografted hepatocytes was observed postnatally, potentially due to cytotoxicity from NK cells. Strategies to enhance engraftment and reduce NK cell-mediated rejection are needed for large-scale expansion of human hepatocytes in immunodeficient swine.

Summary: The study found that a common human variant, TNFRSF13B A181E, confers resistance to enteric pathogens in mice, primarily due to natural IgA deficiency in the intestine. In both WT mice and mutant mice reconstituted with enteric IgA from WT mice, IgA induces the expression of virulence genes, leading to pathogenicity and transmission.

Summary: Cold ischemic preservation results in greater dysfunction, cell death, and inflammation in the left ventricle compared to the right ventricle of mouse hearts, with differential expression of inflammatory cytokines and inflammasome components. However, elevated IL-6 protein levels and transcriptional profiles are similar in both ventricles. Similar findings are observed in human donor hearts.
