Summary: Cyclophilins, which are overexpressed in hepatocellular carcinoma (HCC), play a key role in cancer progression and mediating chemoresistance. The novel cyclophilin inhibitor NV651 can reduce cell division and DNA repair in HCC cells. Combination treatment of NV651 and cisplatin significantly increases cancer cell death compared to cisplatin alone.

Summary: This study found that cordycepin weakens the function of M1-like macrophages in the tumor microenvironment and contributes to macrophage polarization towards the M2 phenotype. Combination treatment with cordycepin and an anti-CD47 antibody enhances the tumor-suppressive effect of cordycepin and reactivates macrophages. It also regulates the proportion of CD8(+) T cells and prolongs the progression-free survival of patients with digestive tract malignancies.

Summary: This study investigated the antitumor active components and synergistic mechanism of Portulaca oleracea L. (PO), an edible plant and traditional medicine, on cervical cancer using network pharmacology and experimental validation. The results showed that combining PO active components (POF) with cisplatin had a stronger anti-tumor effect. Flavonoids were identified as core components, and POF and cisplatin shared common targets related to apoptosis in cervical cancer. The mechanisms involved inducing apoptosis through the PI3K/Akt signaling pathway. This study revealed the key ingredients, potential targets, and mechanism of PO combined with cisplatin against cervical cancer.

Summary: Oral cancer has traditionally been treated with surgery, radiotherapy, chemotherapy, or a combination of these therapies. Recent studies have shown that 3,5-Bis (4-hydroxy-3-methoxybenzylidene)-N-methyl-4-piperidine (PAC), a new curcumin analog, possesses anticancer properties and could be considered a complementary or alternative therapy. The combination of PAC and cisplatin enhances oral cancer cell death by inducing apoptosis, autophagy, and oxidative stress, indicating PAC's potential as a powerful complementary agent to cisplatin in the treatment of gingival squamous cell carcinomas.

Summary: The combination therapy of berberine and cisplatin significantly inhibited osteosarcoma cell proliferation, colony formation, migration, invasion, induced apoptosis, and cell cycle arrest in the G0/G1 phase. Mechanistically, the combination treatment downregulated the expression of MMP-2/9, Bcl-2, CyclinD1, and CDK4, upregulated Bax expression, and regulated the activity of the MAPK pathway.

Summary: This study found that cordycepin, an extract from Cordyceps militaris, has a significant protective effect on glucotoxicity and lipotoxicity-induced damage in pancreatic islet cells. It improves cell viability, energy metabolism, and insulin synthesis and secretion. The mechanism may involve reducing ROS levels, increasing ATP content, causing membrane depolarization, balancing Ca2+ concentration, and inhibiting cell apoptosis through downregulation of the JNK pathway.

Summary: This study aimed to investigate the combined efficacy of Compound Kushen Injection (CKI)-derived combination strategies in p53-mutant CRC. The results showed that CKI combined with cisplatin (Cis) produced synergistic antitumor efficiency for p53-R273H/P309S mutant CRC cells. The combination of CKI and Cis reinforced cell death in a synergistic manner by inducing extrinsic apoptosis pathway.

Summary: In this study, the feasibility and mechanism of cordycepin combined with doxorubicin for liver cancer treatment were investigated. Both in vitro and in vivo studies demonstrated that the combination effectively inhibited cell proliferation and induced apoptosis in hepatocellular carcinoma. Network pharmacology analysis revealed the potential targets and pathways involved in the anticancer mechanism. Additionally, the combination treatment regulated the expression of genes related to cell migration, thus inhibiting cell migration.

Summary: This study identified a new late-stage autophagy inhibitor called CUR5g, which selectively inhibited autophagosome degradation in cancer cells. CUR5g showed potential for the treatment of non-small-cell lung cancer (NSCLC), particularly when combined with cisplatin, as it enhanced cisplatin sensitivity and effectively inhibited autophagy in tumor tissues.

Summary: This study investigated the combined effects and apoptotic mechanisms of hyperthermia and cordycepin on human leukemia U937 cells. The results showed that combined treatment with hyperthermia and cordycepin significantly augmented apoptosis by upregulating Bax and suppressing Bcl-2, Bid, and activated caspase expression. The MAPK pathway and mitochondrial dysfunction were also involved in the induction of apoptosis. Furthermore, NAC could rescue the cells by restoring ROS levels and caspase-3 activity. Cordycepin has the potential to be used as a hyperthermia sensitizer to treat leukemia.

Summary: The study showed that Cor exacerbates Cd-induced cognitive impairment and neurotoxicity in mice, induces synergistic growth inhibition and promotes apoptosis of SHSY5Y cells through the activation of endoplasmic reticulum (ER) stress. This suggests that Cor could enhance Cd-induced neurotoxicity by promoting ER stress-associated apoptosis, leading to potential implications for consumer safety and detection criteria for Cd in Cordyceps militaris.

Summary: The study demonstrates that the combination of cordycepin (Cor) and cisplatin (DDP) has a synergistic effect on inhibiting proliferation and promoting apoptosis of non-small cell lung cancer (NSCLC) cells. The synergistic actions are associated with the activation of AMPK and inhibition of the AKT pathway. This combination therapy may offer a new therapeutic option for DDP-resistant NSCLC.
