Ionic liquids (ILs) are a fascinating group of new chemicals with the potential to replace the classical volatile organic solvents, stimulating many applications in chemical industry. In case ILs are released to the environment, possible combined toxicity should be taken into account and it is, however, often neglected up to now. In this paper, therefore, the concentration-response curves (CRCs) of four groups of IL mixtures with various mixture ratios to Vibrio qinghaiensis sp.-Q67 were determined using the microplate toxicity analysis and were compared to the CRCs predicted by an additive reference model, the concentration addition (CA) or independent action (IA), to identify the toxicity interaction. It is showed that most of the IL mixture rays displayed the classical addition while the remaining rays exhibited antagonism or synergism. Moreover, it is found that the pEC(50) values of the mixture rays exhibiting antagonism or synergism are well correlated with the mixture ratio of a certain IL therein. New methods of top-to-down mixture toxicity prediction: A case study of eliminating of the effects of cosolvent from binary mixtures

Ze-Jun Wang, Qiao-Feng Zheng, Shu-Shen Liu, Peng Huang, Ting-Ting Ding, Ya-Qian Xu

Summary: This study focuses on predicting mixture toxicity based on single substances or low component mixtures using concentration addition (CA) and independent action (IA) models. Two top-to-down methods, inverse CA (ICA) and inverse IA (IIA) models, were proposed to eliminate the effects of specific components from mixtures. The results showed that cosolvent IPA with hormesis may cause unpredictable effects, while DMSO effectively eliminates the effects of toxic components from mixtures. These methods provide a new framework for mixture toxicity evaluation and risk assessment.

Hormetic dose-response of halogenated organic pollutants on Microcystis aeruginosa: Joint toxic action and mechanism

Yueheng Zhang, Qing Gao, Shu-shen Liu, Liang Tang, Xin-Gui Li, Haoyu Sun

Summary: Quinolones, dechloranes, and chlorinated paraffins are new types of halogenated organic pollutants that come from flame retardants, lubricants, and pesticides. This study investigates the toxic effects of these pollutants on cyanobacteria, particularly Microcystis aeruginosa, and finds that quinolones and dechloranes have hormetic effects on their proliferation, while chlorinated paraffins do not. The joint toxicity tests show that mixtures of quinolones & chlorinated paraffins and dechloranes & chlorinated paraffins also exhibit hormetic effects on Microcystis aeruginosa. However, mixtures of quinolones & dechloranes do not have a hormetic effect. It is also found that mixtures of quinolones & dechloranes have synergistic toxic actions, while the joint toxic actions of both quinolones & chlorinated paraffins, and dechloranes & chlorinated paraffins shift from addition to antagonism with increasing concentration. The study provides important data and mechanisms for understanding the hormetic phenomenon of these pollutants on Microcystis aeruginosa and highlights the role of hormesis in the risk assessment of environmental pollutants.

Decomposition of Total Organic Halogen Formed during Chlorination: The Iceberg of Halogenated Disinfection Byproducts Was Previously Underestimated

Chao Fang, Xinmiao Luan, Feiyang Ao, Xingyu Wang, Shunke Ding, Zhenqi Du, Shushen Liu, Ruibao Jia, Wenhai Chu

Summary: In this study, it was discovered that the level of TOX in chlorinated waters had been significantly underestimated under common analytical conditions. The addition of quenching agent sodium thiosulfate resulted in a two-phase decomposition pattern of total organic chlorine and total organic bromine, indicating the important toxicological significance of the DBPs responsible for TOX decomposition. Nontarget analysis using high-resolution mass spectrometry revealed the molecular formulas for the decomposed TOX, and it was found that the level of unknown TOX in chlorinated waters was considerably higher than previously thought.

Transfer pattern of hormesis into personal care product mixtures from typical hormesis-inducing compounds

Ya-Qian Xu, Kai Li, Ze-Jun Wang, Peng Huang, Shu-Shen Liu

Summary: This study investigates the transfer of hormesis between chemical components and personal care product mixtures and identifies a key compound, betaine, affecting hormesis transfer. The study finds that only mixture rays containing at least one hormesis-inducing compound produce hormesis to the freshwater photobacterium Q67. The study also suggests that betaine triggers the quorum sensing pathway, contributing to hormetic effects. This research provides new insights into hormesis and its mixtures, and offers an efficient method to identify potential components that affect hormesis transfer in mixtures.

Study on the characterization of pesticide modes of action similarity and the multi-endpoint combined toxicity of pesticide mixtures to Caenorhabditis elegans

Peng Huang, Shu-Shen Liu, Ze-Jun Wang, Ting-Ting Ding, Meng-Ting Tao, Zhong-Wei Gu

Summary: With the widespread use of pesticides, the cocktail effect caused by the coexistence of multiple low-residue pesticides in the environment has become a concern. However, limited information on the modes of action (MOAs) of chemicals hinders the application of concentration addition (CA) models in evaluating and predicting the toxicity of mixtures with similar MOAs. This study characterized the similarity of pesticide MOAs using MEDV-13 descriptors and established methods to test the toxicity of mixtures on the lifespan and reproduction of Caenorhabditis elegans. The results showed consistent toxicity interactions of the mixtures on the nematode's lifespan and reproductive endpoints.

Optimized Derivation of Predicted No-Effect Concentrations (PNECs) for Eight Polycyclic Aromatic Hydrocarbons (PAHs) Using HC10 Based on Acute Toxicity Data

Xiao Sun, Ting-Ting Ding, Ze-Jun Wang, Peng Huang, Shu-Shen Liu

Summary: In this study, the application of the species sensitivity distribution (SSD) for ecological risk assessment of persistent organic pollutants (POPs), specifically polycyclic aromatic hydrocarbons (PAHs), is improved by exploring the use of acute SSD curves to derive chronic predicted no-effect concentrations (PNECs) and calculating PNECs using HC10 instead of HC5. The results show that the chronic PNEC for PAHs can be estimated by multiplying the acute PNEC by 0.1 and the assessment factor for HC10 is 10. The obtained chronic PNECs based on acute HC10 values for specific PAHs are reported.

A novel mixture sampling strategy combining latin hypercube sampling with optimized one factor at a time method: A case study on mixtures of antibiotics and pesticides

Ting-Ting Ding, Shu-Shen Liu, Ze-Jun Wang, Peng Huang, Meng-Ting Tao, Zhong-Wei Gu

Summary: Global sensitivity analysis combined with quantitative high-throughput screening provides a novel technique for identifying the key components responsible for the toxicities of mixtures. However, the current designs suffer from uneven frequency sampling, repeated mixtures, and only considering odd factor levels. To address these issues, the study proposes a method called LHS-OAT, which uses latin hypercube sampling to achieve equal frequency sampling and non-repeated mixtures, and applies different one factor at a time methods for factors with odd and even levels. The method was successfully applied to design mixtures of antibiotics and pesticides, accurately identifying the important factors inducing the toxicities of the mixtures.
