Summary: Certain supramolecular motifs, such as Cucurbit[7]uril (CB[7]) macrocycles, exhibit affinities comparable to well-known biological interactions. By modifying cells with CB[7] constructed from a polyamidoamine (PAMAM) dendrimer scaffold, guest-linked drugs can be delivered to the cell membrane, offering potential applications in biomedicine. This approach has the potential to be used for in situ imaging, modulation of cell-based therapies, or the construction of synthetic supramolecular recognition axis on the cell membrane.

Summary: The study found that the self-assembly of synthesized macrocycles is mainly driven by hydrophobic interactions of their hydrophobic chains and π-π stacking interactions between aromatic rings. Long-chain viologen macrocycles not only have low hemotoxicity and selective cytotoxic activity, but also can bind therapeutic molecules, making them promising drug nanocontainers.

Summary: The discovery of lipid-hybrid nanosystems has provided potential solutions to drug delivery and theranostic challenges. However, the commonly used components in these systems often pose challenges related to solubility, physicochemical properties, immune compatibility, and tunability. In this study, a new supramolecular scaffold with synthetic tunability and amphiphilic characteristics was designed and synthesized. The scaffold was found to form stable blended nanospheres with sustained drug release properties. In vitro and in vivo assays demonstrated the efficacy of these nanosystems in drug encapsulation and delivery.
