Summary: This study investigated the expression of GPBAR1 in HCC tissues and tumor-adjacent tissues and found that GPBAR1 was highly expressed in HCC tissues. GPBAR1 expression was significantly associated with poor prognosis in HCC and served as an independent prognostic biomarker. Additionally, GPBAR1 expression was significantly correlated with asynchronous bone metastasis but not liver or lung metastases. These findings provide new insights into HCC metastasis and improve prognostic assessment of HCC.

Summary: Cutaneous metastases from hepatocellular carcinoma (HCC) as an extrahepatic metastasis are rare and have a poor prognosis. This study summarized 12 cases of non-iatrogenic implantation of cutaneous metastasis after HCC, and investigated the clinical prognosis, pathological characteristics, and treatment. The findings suggest that systematic treatment is recommended for large HCC to prevent cutaneous metastasis and improve prognosis.

Summary: In this study, hepatocellular carcinoma (HCC)-associated anoikis-related genes (ANRGs) were identified and a robust prognostic model for HCC was established based on the ANRGs. The study also characterized the immune characteristics based on the anoikis signature. The results provide a valuable tool for clinicians in selecting targeted therapeutic drugs and understanding the mechanisms underlying immune cell infiltration regulated by anoikis.

Summary: This study aimed to investigate the risk and survival of synchronous and metachronous bone metastasis in hepatocellular carcinoma (HCC), and to reveal the characteristics and related factors of HCC patients with bone metastasis. The results showed that the incidence of synchronous bone metastasis in HCC was 1.74% at initial diagnosis, and the median latency time from HCC diagnosis to metachronous bone metastasis was 6 months. Alcohol-drinking history, number and size of intrahepatic tumor, lymph node metastasis, serum ALP and HGB level were found to be correlated with the occurrence of bone metastasis. The overall survival between non-bone metastasis and bone metastasis patients were statistically different.

Summary: Liver cancer is a major cause of cancer-related death globally, with limited therapeutic options available. Reactivation of receptor tyrosine kinases (RTK) has been identified as a mechanism of resistance to targeted therapies in various cancers. In this study, we demonstrate that multiple RTK are activated upon mTOR inhibition in hepatocellular carcinoma (HCC), leading to reactivation of the mTOR pathway. Co-inhibition of mTOR and SHP2, a downstream target of RTK, synergistically induces apoptosis and effectively impairs tumor growth in HCC mouse models. These findings suggest a novel rational combination therapy for HCC.

Summary: Tumor-associated macrophages (TAMs) promote the activation of hepatic leukemia factor (HLF) in triple-negative breast cancer (TNBC) cells through the secretion of transforming growth factor-beta1 (TGF-beta 1) and subsequently, HLF transactivates gamma-glutamyltransferase 1 (GGT1) to enhance ferroptosis resistance. This process drives TNBC cell proliferation, metastasis, and cisplatin resistance. Additionally, TNBC cells activate the JAK2/STAT3 axis to induce TGF-beta 1 secretion by TAMs, forming a feed-forward circuit. The combination of HLF and GGT1 values can improve the accuracy of TNBC patient prognosis.

Additionally, GSEA analysis revealed enrichment of immune-related pathways in the high-risk group, and ssGSEA analysis showed increased infiltration of immune cells and activities of immune pathways in the same group.
