Synthesis, characterisation and cytotoxic activity evaluation of new metal-salen complexes based on the 1,2-bicyclo[2.2.2]octane bridge

Pierre Milbeo, Francois Quintin, Laure Moulat, Claude Didierjean, Jean Martinez, Xavier Bantreil, Monique Calmes, Frederic Lamaty

Summary: (R)-1,2-Diaminobicyclo[2.2.2]octane was used to prepare a salen ligand, and five metal salen complexes were synthesized with high yield. The original manganese salen complex showed the highest cytotoxic activity, being 16 times more potent than cisplatin, highlighting the advantage of the bridging backbone. An alternative mechanochemical method for preparing this complex was also explored.

Synthesis of α-Amino Acid N-Carboxyanhydrides

Guillaume Laconde, Muriel Amblard, Jean Martinez

Summary: A simple method for synthesizing α-amino acid N-carboxyanhydrides (NCAs) without using phosgene and halogen is described. The process is safe, easy to operate, and does not require special installation, generating nontoxic, easy to remove byproducts. It is suitable for on-demand on-site production of NCAs in small or large quantities.

Controlled Silylation of Polysaccharides: Attractive Building Blocks for Biocompatible Foams and Cell-Laden Hydrogels

Titouan Montheil, Laurine Raimond, Laurine Valot, Marie Maumus, Matthieu Simon, Jean Martinez, Muriel Amblard, Daniele Noel, Ahmad Mehdi, Gilles Subra

Summary: We have developed a general methodology to synthesize and isolate hybrid silylated polysaccharides, which can be used to prepare cell-laden hydrogels through sol-gel polymerization in aqueous buffer and can be transformed into hybrid xerogels by freeze-drying. These materials have the potential to be used as biocompatible foams for biological applications.

The C-terminal pentapeptide acein analogue (JMV3315) stimulates dopamine release in the brain

Charlene Lucas-Valmalle, Gilles Subra, Pascal Verdie, Marie-Lou Kemel, Valerie Dauge, Karine Puget, Jean Martinez

Summary: This study investigated the structure-activity relationships of acein by conducting displacement experiments using guinea pig brain membranes with high-affinity acein binding sites, using the labeled ligand [I-125]Tyr-Pro-Pro-Thr-Thr-Thr-Lys-Phe-Ala-Ala-OH. The results showed that the C-terminal pentapeptide H-Thr-Lys-Phe-Ala-Ala-OH is the minimal structure with high affinity for acein binding sites. Among the synthesized analogues, the pentapeptide H-Thr-Lys-Tyr-Ala-Ala-OH exhibited the highest binding affinity.

Encapsulation of BSA in hybrid PEG hydrogels: stability and controlled release

Corine Tourne-Peteilh, Maeva Barege, Mathieu Lions, Jean Martinez, Jean-Marie Devoisselle, Anne Aubert-Pouessel, Gilles Subra, Ahmad Mehdi

Summary: Hybrid hydrogels based on silylated polyethylene glycol (Si-PEG) were evaluated as matrices for controlled release of bovine serum albumin (BSA), with investigations into the parameters affecting the hydrogel structure and protein release behavior. The study provided insights into the stability, swelling, and mechanical properties of the network, as well as the impact of different factors on the protein release profiles.

A Collagen-Mimetic Organic-Inorganic Hydrogel for Cartilage Engineering

Laurine Valot, Marie Maumus, Luc Brunel, Jean Martinez, Muriel Amblard, Daniele Noel, Ahmad Mehdi, Gilles Subra

Summary: Researchers have developed a biomimetic chemical hydrogel based on silylated collagen-mimetic synthetic peptides, which successfully encapsulated MSCs and improved mechanical properties, resulting in prolonged survival of the embedded cells and enhanced expression of chondrocyte markers. This long-standing hybrid hydrogel shows promise as a synthetic and modular scaffold for cartilage tissue engineering.
