The head-to-tail conversion of linear arylopeptoids (oligomeric N-substituted aminomethyl benzamides) into the derived novel macro-cycles has enabled the first X-ray structures of arylopeptoid constructs and the identification of well-defined architectures in solution.

First series of N-alkylamino peptoid homooligomers: solution phase synthesis and conformational investigation

Summary: The synthesis and conformational analysis of peptoid oligomers composed of consecutive N-(alkylamino)glycine units were investigated. N-(Methylamino)glycine homooligomers were successfully synthesized in solution and their structures were analyzed using various techniques. It was found that N-(methylamino)peptoids preferentially adopt trans amide bonds and can form intermolecular associations via hydrogen bonding.

This series provides a promising basis for further research.

Biochemical and Structural Insights on the Poplar Tau Glutathione Transferase GSTU19 and 20 Paralogs Binding Flavonoids

Summary: This study focuses on the expression and function of glutathione transferases (GSTs) in Populus trichocarpa. The results reveal that the tau class (GSTU) is the most abundant in this species. The study also identifies a common ancestor for PtGSTU19 and PtGSTU20 genes and highlights the structural differences between them. Interaction with flavonoids is found to potentially play a role in the GSH-conjugation activity of these two genes.

Synthesis and Kinase Inhibitory Potencies of Pyrazolo[3,4-g]isoquinolines

Summary: A new series of pyrazolo[3,4-g]isoquinoline derivatives, diverse in substitution at the 4- or 8-position, were synthesized and evaluated for their kinase inhibitory potency. The results showed that the introduction of a bromine atom at the 8-position had a negative effect on Haspin inhibition, while the introduction of an alkyl group at the 4-position led to a modification of the kinase inhibition profiles. Overall, the new pyrazolo[3,4-g]isoquinolines represent a novel family of kinase inhibitors with various selectivity profiles.

Summary: We developed dipeptidomimetic compounds as potent inhibitors of nNOS and iNOS, showing significant selectivity against eNOS. The extension size was increased to improve interactions in the NOS substrate access channel. Molecular modeling explained the selectivity towards nNOS over eNOS, and spectral studies confirmed interaction at the heme active site. These selected inhibitors were effective in inhibiting intra-cellular nNOS and iNOS in RAW264.7 and INS-1 cells.

Summary: Neuropilin 1 (NRP1), a cell-surface co-receptor, has been implicated in the development and progression of various cancers. Researchers have utilized a combination of the KDKPPR peptide, a photosensitizer, and a contrast agent to target NRP1 and treat glioblastoma. By mutating specific amino acids on the protein surface, crystallography was used to determine the structure of the mutated fragment and its complex with the KDKPPR peptide, providing insights into the electrostatic interactions involved in ligand binding. Molecular dynamics simulations further analyzed the binding and mobility of the wild-type and mutant proteins.

Unveiling the conformational landscape of achiral all-cis tert-butyl β-peptoids

Summary: The synthesis and conformational study of N-substituted beta-alanines with tert-butyl side chains are described in this article. Experimental solution NMR spectroscopy, X-ray crystallography, and molecular modeling were used to investigate the conformational preferences of these compounds. It was found that despite their higher flexibility, they still adopt preferred secondary structures including helical and ribbon-like conformations.

Structural Insights into a Fusion Protein between a Glutaredoxin-like and a Ferredoxin-Disulfide Reductase Domain from an Extremophile Bacterium

Summary: This study reveals the three-dimensional structure of a natural hybrid protein formed by the fusion of an N-terminal glutaredoxin-like domain and an FDR domain in the marine bacterium Desulfotalea psychrophila Lsv54. The structure provides insights into the dispensability of the variable subunit that normally protects the Fe-S cluster in FDR proteins and the electron flow involving RBX, FDR, and GRX protein domains observed in anaerobic methanogen archaea.

Elucidating the effect of the spacer and the luminescence mechanism of SRB hosted in a LDH interlayer

Summary: The study investigates the optical properties of sulforhodamine B hosted in surfactant-modified layered double hydroxide, examining the effects of different spacers on the luminescence of the optically active molecules. It demonstrates that intercalation of spacers allows for adjustment of distance between LDH sheets based on the size of the luminescent molecule guest, and identifies the surfactant with n = 11 as providing the best performance in terms of optical properties. The use of computational approaches and silicon composite films with LDH loadings further reveal insights into the distribution and effects of different alkyl chain lengths on the luminescent molecule within the LDH interlayer.

Pd/Brønsted acid catalysed intramolecular N-allylation of indoles and pyrroles with alkynes for the synthesis of N-fused heterocycles

Summary: In this article, a novel catalytic reaction using Pd(0) and Bronsted acid is reported for the synthesis of biologically important imidazolidinone-fused N-heterocycles through redox-neutral intramolecular N-allylation of indoles and pyrroles with alkynes. This atom-economical method is applicable to a wide range of substrates and eliminates the need for leaving groups or oxidizing agents commonly used in traditional allylation reactions.

Rh(III)-catalyzed selective mono- and dual-functionalization/cyclization of 1-aryl-5-aminopyrazoles with iodonium ylides

Modified minimal-size fragments of heparan sulfate as inhibitors of endosulfatase-2 (Sulf-2)

Summary: This study presents the design and synthesis of sulfated disaccharide inhibitors based on IdoA(2S)-GlcNS(6S), which showed potent inhibition of Sulf-2. The results suggest that IdoA(2S)-GlcNS(6S) is the shortest fragment size required for effective inhibition of Sulfs, and a trisulfated disaccharide is identified as the minimal fragment size of heparan sulfate for effective endosulfatase inhibition.

Reductive cyclodimerization of chalcones: exploring the self-adaptability of galvanostatic electrosynthesis

Summary: The self-adaptability of galvanostatic electrolysis was found to be helpful in a multistage chemo- and diastereoselective electrochemically promoted cyclodimerization of chalcones. Through a series of reductive events, densely functionalized cyclopentanes with five contiguous stereocenters were obtained (25 examples, yields up to 95%, dr values up to >20:1). Further experimental and electrochemical investigations indicated the crucial role of dynamic kinetic resolution of the aldol intermediate in the reaction mechanism.

Rh(III)-catalyzed oxidative [4+2] annulation of 2-arylquinoxalines and 2-aryl-2H-indazoles with allyl alcohols

Summary: In this study, the synthesis of functionalized benzo[a]phenazines and indazolo[2,3-a]quinolines was achieved through Rh(III)-catalyzed oxidative annulation of 2-arylquinoxalines and 2-aryl-2H-indazoles with allyl alcohols. The method showed a wide substrate scope, excellent functional group tolerance, good to high yields of annulated products, and the capability for scaled-up synthesis. A tentative mechanism of the annulation reaction was proposed based on a preliminary mechanistic investigation.
