Summary: The development of new methods for introducing fluorine atoms or fluorinated moieties in molecular scaffolds is crucial in organic synthesis. The incorporation of the SCF3 group into organic molecules has shown beneficial effects on drug stability and bioavailability. By using a chiral auxiliary, enantioenriched alpha-SCF3-tetrasubstitued beta-keto esters can be synthesized in good yields with high enantioselectivity.

Summary: This study introduces a novel strategy for the enantioselective synthesis of axially chiral N,N'-bisindoles using chiral phosphoric acid-catalyzed formal cycloadditions. The synthesized compounds showed excellent yields and enantioselectivities, and exhibited cytotoxicity towards cancer cells. Moreover, they were also transformed into axially chiral phosphine ligands with high catalytic activity.

Summary: This paper summarizes our recent efforts in the enantioselective organocatalytic synthesis of chiral halogenated compounds. The discussion includes enantioselective alpha-halogenation of aldehydes, decarboxylative chlorination of beta-keto acids, and enantioselective C-C bond formation at the trifluoromethylated prochiral carbon to yield organohalides with chlorinated, fluorinated, or trifluoromethylated chiral stereogenic centers. Common organocatalysts such as Jorgensen-Hayashi catalyst and cinchona alkaloid-derivatived catalyst were applied, and novel chiral amine catalysts were developed for these reactions. Stereospecific derivatizations of the chiral halogenated compounds via nucleophilic substitution are also discussed, resulting in the synthesis of many novel chiral compounds that have not been reported before, even as racemates.

Summary: Efficient and practical methodology has been developed for building two types of phosphorus-installing helicenes via the phosphonium salt catalyzed phospha-Michael reaction together with copper salt enabled aromatization. The scalability and practicality of this protocol were validated by the gram-scale synthesis and transformation of products, particularly towards producing novel helical phosphine ligands that were successfully utilized in metal-catalyzed reactions.

Summary: We developed a simple and practical protocol for the chemoselective coupling of optically active functionally rich 2-aroylcyclopropanecarbaldehydes with various CH acids or active methylene compounds. This metal-free, organocatalytic selective reductive C-C coupling method offers advantages such as no epimerization, no ring opening, high carbonyl control, and large substrate scope. The resulting chiral products can serve as building blocks for the synthesis of value-added small molecules, natural products, pharmaceuticals, and their analogues.

Summary: Enantioconvergent iso-Pictet-Spengler reactions of chiral racemic ss-formyl esters and a ss-keto ester are reported, leading to the formation of complex tetrahydro-& gamma;-carbolines bearing two contiguous stereocenters. These reactions are catalyzed by a chiral thiourea and benzoic acid cocatalytic system, representing unusual nonhydrogenative stereoconvergent additions to racemic & alpha;-stereogenic-ss-dicarbonyls. The transformation of the products to chiral aminoalcohols and carbamates is also demonstrated.

Summary: This study presents the design and synthesis of sulfated disaccharide inhibitors based on IdoA(2S)-GlcNS(6S), which showed potent inhibition of Sulf-2. The results suggest that IdoA(2S)-GlcNS(6S) is the shortest fragment size required for effective inhibition of Sulfs, and a trisulfated disaccharide is identified as the minimal fragment size of heparan sulfate for effective endosulfatase inhibition.

Summary: The self-adaptability of galvanostatic electrolysis was found to be helpful in a multistage chemo- and diastereoselective electrochemically promoted cyclodimerization of chalcones. Through a series of reductive events, densely functionalized cyclopentanes with five contiguous stereocenters were obtained (25 examples, yields up to 95%, dr values up to >20:1). Further experimental and electrochemical investigations indicated the crucial role of dynamic kinetic resolution of the aldol intermediate in the reaction mechanism.

Summary: In this study, the synthesis of functionalized benzo[a]phenazines and indazolo[2,3-a]quinolines was achieved through Rh(III)-catalyzed oxidative annulation of 2-arylquinoxalines and 2-aryl-2H-indazoles with allyl alcohols. The method showed a wide substrate scope, excellent functional group tolerance, good to high yields of annulated products, and the capability for scaled-up synthesis. A tentative mechanism of the annulation reaction was proposed based on a preliminary mechanistic investigation.
