4 alpha- Aminosteroids were synthesized by the substitution of a 2 alpha-bromo ketone using K2CO3 as an activator; 4 beta-aminosteroids were synthesized in excellent yields by a highly regioselective and trans-stereospecific ring opening of a steroidal 3,4a- epoxide using ZnCl2-H2O as a catalyst.

Summary: This study utilized mass spectrometry-based quantitative proteomic strategy to investigate the molecular differences in HDACi vorinostat (SAHA) sensitive and resistant cell lines. The results revealed potential new drug combinations and shed light on the molecular mechanisms of drug resistance and therapeutic efficacy in solid malignancies. Proteomic analysis indicated alterations in the transcription process and protein homeostasis may contribute to the synergistic utilization of drug combinations.

Summary: The study developed a light-induced lysine (K) enabled crosslinking (LIKE-XL) strategy for spatiotemporal and global profiling of DNA-protein interactions. By capturing weak and transient DNA-protein interactions, LIKE-XL can discover low-affinity transcription-factor/DNA interactions using specific DNA baits and identify previously unknown binding sites. Additionally, it can decipher the dynamics of the transcription factor subproteome in response to drug treatment and identify downstream target transcription factors, providing insight into their dynamic transcriptional networks. LIKE-XL offers a complementary method to expand the DNA-protein profiling toolbox and map accurate DNA-protein interactomes for better understanding of protein function in health and disease.

Summary: AMPK inhibits lung cancer metastasis by relieving H3K9me2-mediated silencing of epithelial genes (e.g., CDH1) through activation by metformin. This process involves interaction between PHF2, a H3K9me2 demethylase, and AMPK alpha 2. AMPK phosphorylates PHF2 at S655 site, enhancing PHF2 demethylation activity and promoting the transcription of CDH1.

Pd/Brønsted acid catalysed intramolecular N-allylation of indoles and pyrroles with alkynes for the synthesis of N-fused heterocycles

Summary: In this article, a novel catalytic reaction using Pd(0) and Bronsted acid is reported for the synthesis of biologically important imidazolidinone-fused N-heterocycles through redox-neutral intramolecular N-allylation of indoles and pyrroles with alkynes. This atom-economical method is applicable to a wide range of substrates and eliminates the need for leaving groups or oxidizing agents commonly used in traditional allylation reactions.

Modified minimal-size fragments of heparan sulfate as inhibitors of endosulfatase-2 (Sulf-2)

Summary: This study presents the design and synthesis of sulfated disaccharide inhibitors based on IdoA(2S)-GlcNS(6S), which showed potent inhibition of Sulf-2. The results suggest that IdoA(2S)-GlcNS(6S) is the shortest fragment size required for effective inhibition of Sulfs, and a trisulfated disaccharide is identified as the minimal fragment size of heparan sulfate for effective endosulfatase inhibition.

Reductive cyclodimerization of chalcones: exploring the self-adaptability of galvanostatic electrosynthesis

Summary: The self-adaptability of galvanostatic electrolysis was found to be helpful in a multistage chemo- and diastereoselective electrochemically promoted cyclodimerization of chalcones. Through a series of reductive events, densely functionalized cyclopentanes with five contiguous stereocenters were obtained (25 examples, yields up to 95%, dr values up to >20:1). Further experimental and electrochemical investigations indicated the crucial role of dynamic kinetic resolution of the aldol intermediate in the reaction mechanism.

Rh(III)-catalyzed oxidative [4+2] annulation of 2-arylquinoxalines and 2-aryl-2H-indazoles with allyl alcohols

Summary: In this study, the synthesis of functionalized benzo[a]phenazines and indazolo[2,3-a]quinolines was achieved through Rh(III)-catalyzed oxidative annulation of 2-arylquinoxalines and 2-aryl-2H-indazoles with allyl alcohols. The method showed a wide substrate scope, excellent functional group tolerance, good to high yields of annulated products, and the capability for scaled-up synthesis. A tentative mechanism of the annulation reaction was proposed based on a preliminary mechanistic investigation.
