Summary: Hybrid peptides composed of alpha and beta-amino acids have potential as therapeutic agents. Therefore, the development of new unnatural amino acids and methods for their incorporation is important. In this study, we investigated the application of 5-amino-3-methyl-isoxazole-4-carboxylic acid (AMIA) in solid phase peptide synthesis. The results show that this unnatural beta-amino acid can be used in synthesizing a new class of bioactive peptides.

Summary: This article presents an alternative practical method for the asymmetric synthesis of beta-amino-alpha-ketoacid equivalents, utilizing a highly stereoselective organocatalyzed Mannich-type addition using glyoxylate cyanohydrin. The optimal aminothiourea catalyst provides a variety of adducts with excellent yield and stereoselectivity, and the reaction can be applied to a wider range of imine precursors. Experimental and computational studies reveal the factors contributing to the high diastereoselectivity and enantioselectivity of the reaction.

Summary: Well-defined absorption spectra of well-characterised alpha-glycine were obtained in the spectral range 2.5-7.5 THz, with four separate resonances observed at 93, 152, 188, and 223 cm(-1) at the lowest temperature. The observed frequency reduction with increasing temperature is attributed to phonon-mediated anharmonicity in the crystal potential.

Summary: In this study, the activity of TxID and its loop2-modified mutants were evaluated using an electrophysiological assay. The results showed a decrease in inhibition against r alpha 3 beta 4 and r f alpha 6/ alpha 3 beta 4 nAChRs for the 4/7-subfamily mutants [+9A]TxID, [+10A]TxID, [+14A]TxID, and all the 4/5-subfamily mutants, indicating that the insertion or truncation of the 9th, 10th, and 11th amino acid results in a loss of inhibition, and the truncation of loop2 has more pronounced impacts on its functions. These findings enhance the understanding of ff-conotoxin, provide guidance for further modifications, and offer a perspective for future studies on the molecular mechanism of the interaction between ff-conotoxins and nAChRs.

The mutant Y314R demonstrated higher diastereomeric excess (de) in asymmetric synthesis, showcasing the potential of directly controlling substrates in the active pocket for industrial applications.

Summary: The study developed a light-induced lysine (K) enabled crosslinking (LIKE-XL) strategy for spatiotemporal and global profiling of DNA-protein interactions. By capturing weak and transient DNA-protein interactions, LIKE-XL can discover low-affinity transcription-factor/DNA interactions using specific DNA baits and identify previously unknown binding sites. Additionally, it can decipher the dynamics of the transcription factor subproteome in response to drug treatment and identify downstream target transcription factors, providing insight into their dynamic transcriptional networks. LIKE-XL offers a complementary method to expand the DNA-protein profiling toolbox and map accurate DNA-protein interactomes for better understanding of protein function in health and disease.

Summary: In this article, a novel catalytic reaction using Pd(0) and Bronsted acid is reported for the synthesis of biologically important imidazolidinone-fused N-heterocycles through redox-neutral intramolecular N-allylation of indoles and pyrroles with alkynes. This atom-economical method is applicable to a wide range of substrates and eliminates the need for leaving groups or oxidizing agents commonly used in traditional allylation reactions.

Summary: This study presents the design and synthesis of sulfated disaccharide inhibitors based on IdoA(2S)-GlcNS(6S), which showed potent inhibition of Sulf-2. The results suggest that IdoA(2S)-GlcNS(6S) is the shortest fragment size required for effective inhibition of Sulfs, and a trisulfated disaccharide is identified as the minimal fragment size of heparan sulfate for effective endosulfatase inhibition.

Summary: The self-adaptability of galvanostatic electrolysis was found to be helpful in a multistage chemo- and diastereoselective electrochemically promoted cyclodimerization of chalcones. Through a series of reductive events, densely functionalized cyclopentanes with five contiguous stereocenters were obtained (25 examples, yields up to 95%, dr values up to >20:1). Further experimental and electrochemical investigations indicated the crucial role of dynamic kinetic resolution of the aldol intermediate in the reaction mechanism.

Summary: In this study, the synthesis of functionalized benzo[a]phenazines and indazolo[2,3-a]quinolines was achieved through Rh(III)-catalyzed oxidative annulation of 2-arylquinoxalines and 2-aryl-2H-indazoles with allyl alcohols. The method showed a wide substrate scope, excellent functional group tolerance, good to high yields of annulated products, and the capability for scaled-up synthesis. A tentative mechanism of the annulation reaction was proposed based on a preliminary mechanistic investigation.
