Flavusides A (1) and B (2), two new antibacterial cerebroside derivatives, and the previously described phomaligol A (3), kojic acid (4), methyl kojic acid (5), and dimethyl kojic acid (6) have been isolated from the extract of a marine isolate of the fungus Aspergillus flavus. The structure and absolute stereochemistry of two cerebrosides were assigned on the basis of NMR and Tandem FAB-MS/MS experiments. Compounds 1, 2, and 3 exhibited a mild antibacterial activity against Staphylococcus aureus, methicillin-resistant S. aureus, and multidrug-resistant S. aureus. The minimum inhibitory concentration (MIC) values for each strain are as follows: compounds 1 and 2 showed 15.6 mu g/ml for S. aureus and 31.2 mu g/ml for methicillin-resistant S. aureus and multidrug-resistant S. aureus, and compound 3 exhibited 31.2 mu g/ml for S. aureus and methicillin-resistant S. aureus and 62.5 mu g/ml for multidrug-resistant S. aureus.

Taxifolin stability: In silico prediction and in vitro degradation with HPLC-UV/UPLC-ESI-MS monitoring

Fernanda Cristina Stenger Moura, Carmem Lucia dos Santos Machado, Favero Reisdorfer Paula, Angelica Garcia Couto, Maurizio Ricci, Valdir Cechinel-Filho, Tiago J. Bonomini, Louis P. Sandjo, Tania Mari Belle Bresolin

Summary: Taxifolin, isolated from the seeds of Mimusops balata, was found to be extremely unstable under alkaline hydrolysis and its thermal degradation was increased by humidity. However, it displayed stability in terms of photosensitivity. Alkaline degradation products were identified as dimers of taxifolin. These findings contribute to the development of stability-indicating methods and appropriate drug release systems to preserve its stability and enhance bioavailability.

Summary: This study aimed to compare the main plants recognized as arnica in Brazil using metabolomic analysis. The research utilized UPLC-ESI-QTof-MS2 data and multivariate statistical analysis to identify chemical similarities and differences among these plants. The results showed that some species have similar chemical features to Arnica montana, while others are chemically diverse. The study concluded that mass spectrometry combined with multivariate statistical analysis is a valuable tool for identifying correlated compounds and verifying chemical similarity among different plant species.

Summary: Mangroves are unique ecosystems known for their high salinity and organic matter, hosting a rich biodiversity. Fungi, particularly endophytic fungi isolated from mangrove plant species, are an important natural source for discovering new bioactive compounds with potential biotechnological and pharmacological applications. These bioactive compounds show promising bioactivity against protozoa, bacteria, and pathogenic viruses, highlighting the importance of mangrove ecosystems for preservation and as a source of new molecules for controlling pathogens that impact human, animal, and environmental health.

Summary: The hydroalcoholic extract of Polygala altomontana showed a dose-dependent antinociceptive action in the inflammatory phase of the formalin test and anti-hyperalgesic action in a mechanical nociception model. UPLC-ESI-QTOF-MS analysis revealed the presence of phenylpropanoid sucrose esters, glycosylated quercetin derivatives, styrylpyrones, and coumarins in the active extract. Some identified compounds, such as styrylpyrones and coumarins, have demonstrated antinociceptive action. The results suggest the potential of P. altomontana for developing pain-relieving herbal remedies and drugs.

Summary: This study investigates the sensitivity of semisynthetic sesquiterpene lactone (SL) derivatives by preparing them under different pH conditions and evaluating their anti-inflammatory and antiproliferative activities. Results indicate that some of these derivatives exhibit significant cytotoxic activity and selectivity against human melanoma cells. They also demonstrate the ability to reduce the release of interleukin-6 (IL-6) and nitrite (NOx) in pre-treated macrophages. Overall, the study suggests that acidic and alkaline extraction processes can promote the formation of SLs.
