Disruption of the Microtubule Network and Inhibition of VEGFR2 Phosphorylation by Cytotoxic N,O-Coordinated Pt(II) and Ru(II) Complexes of Trimethoxy Aniline-Based Schiff Bases

Summary: Platinum-based and Ruthenium-based complexes have been investigated as potential chemotherapeutic agents with tunable activity against platinum-resistant cancer cells. N,O-coordinated platinum complexes showed superior in vitro antiproliferative activity compared to N,N-coordinated ruthenium complexes, especially against aggressive cancer cells. The complexes also demonstrated disruption of the microtubule network, cell cycle arrest, and apoptotic cell death, indicating their potential for cancer treatment. Among the studied complexes, the platinum-based complexes exhibited higher cytotoxicity, better VEGFR2 inhibition, and strong interaction with a model nucleobase.

Hypoxia Active Platinum(IV) Prodrugs of Orotic Acid Selective to Liver Cancer Cells

Summary: Platinum(IV) complexes of orotic acid have selective targeting towards liver cancer cells, showing enhanced activity and higher uptake in Hep G2 cells. The involvement of OAT2 in the uptake of these complexes is suggested by the higher expression in Hep G2 and decreased toxicity in the presence of an OAT2 inhibitor. These complexes are not sequestered by the copper transporter ATP7B, unlike cisplatin and oxaliplatin.

Selective targeting of the inactive state of hematopoietic cell kinase (Hck) with a stable curcumin derivative

Summary: In this study, a stable Hck inhibitor was developed by functionalizing the 4-arylidene position of the fluorescent curcumin scaffold, showing selective interaction with the inactive conformation of Hck. The lead compound exhibited no inhibitory effects on three other kinases and may mediate cytotoxicity through inhibition of the SFK signaling pathway.

Synthesis, Characterization, and Cytotoxicity of Morpholine-Containing Ruthenium(II) p-Cymene Complexes

However, the low stability of gaseous NO in physiological conditions limits its therapeutic applications, requiring research on more stable NO delivery carriers.
