Summary: By mapping the region-specific nonlinear atrophic trajectory of the whole cerebrum from amnestic mild cognitive impairment (aMCI) to Alzheimer's disease (AD), this study found that the atrophic trajectory is quadratic accelerated, especially in the superior temporal pole, caudate, and hippocampus. Furthermore, the study revealed that APOE ε4 carriers and younger patients had faster atrophic rates in specific regions. Lastly, the identified region-specific biomarkers were able to predict AD conversion within 10 years.

The National Institutes of Health Toolbox Odor Identification Test (NIHTB-OIT) can distinguish between these clinical categories. The scores of NIHTB-OIT decrease with age and are lower for aMCI and ADd participants compared to NC participants.

Summary: Amnestic mild cognitive impairment (aMCI) is the most common preclinical stage of Alzheimer's disease (AD). The study aimed to compare machine learning architectures for classifying and predicting aMCI, assessing the contribution of cognitive measures such as memory binding function in aMCI distinction and prediction. Results indicated that AdaBoost, gradient boosting, and XGBoost had the highest performance in classifying aMCI, while decision tree and random forest had the highest performance in predictive routines.

Summary: This study examined the impact of mild behavioral impairment (MBI) on cognitive performance among cognitively healthy older adults and those with mild cognitive impairment (MCI). The results showed that individuals with MBI performed worse on tasks of attention, episodic memory, executive function, language, and processing speed, and exhibited greater decline over time. The presence of MBI was also associated with poorer performance on tasks of visuospatial ability, executive function, and processing speed among individuals with MCI.

AngleR was the best measure to discriminate HC from tAD (AUC = 0.922), while PerpPD was the best measure to discriminate HC from PCA (AUC = 0.961). Finally, the best global measure to differentiate the two patient groups was ParlPD (AUC = 0.771).

Summary: This research tested a novel in-vivo brain MRI analysis method for investigating cortical architecture changes in patients with Alzheimer's Disease. The method proved to be accurate in detecting altered cortical grey matter quality in AD patients, providing an additional tool for assessing AD and distinguishing patients from healthy controls. Further development of these measurements within the ATN framework may aid in diagnosis, patient selection, and quantification of Neurodegeneration component in response to therapies in clinical trials.
