The Ins and Outs of GluD2-Why and How Purkinje Cells Use the Special Glutamate Receptor

The delta 2 glutamate receptor (GluD2) is predominantly expressed in cerebellar Purkinje cells and plays crucial roles in cerebellar functions. Indeed, the number of synapses between parallel fibers (PFs) and Purkinje cells is specifically and severely reduced in GluD2-null cerebellum. In addition, long-term depression (LTD) at PF-Purkinje cell synapses is impaired in these mice. Nevertheless, the mechanism by which GluD2 regulate these two functions-morphological and functional synaptic plasticity at PF synapses-has remained unclear. Recently, we found that Cbln1, a glycoprotein released from granule cells, was bound to the N-terminal domain of GluD2 and regulated formation and maintenance of PF-Purkinje cell synapses. Furthermore, we found that d-Ser released from Bergmann glia bound the ligand-binding domain of GluD2 and mediated LTD in a manner dependent on the C-terminus. These findings indicate how GluD2 is activated and regulates functions at PF-Purkinje cell synapses. A hypothesis about why GluD2 is employed by PF synapses is also discussed.

Site-specific covalent labeling of His-tag fused proteins with N-acyl-N-alkyl sulfonamide reagent

Summary: In this study, a new reactive peptide tag/probe pair system for site-specific covalent protein labeling was developed. The method relies on the recognition-driven reaction of a peptide tag and a molecular probe. The reactive profile and site-specificity of this method were characterized using model peptides and proteins in vitro, and the general utility for production of a nanobody-chemical probe conjugate without compromising its binding ability was demonstrated.

Subunit-dependent and subunit-independent rules of AMPA receptor trafficking during chemical long-term depression in hippocampal neurons

Summary: The phosphorylation status of GluA1-MPR plays a dominant role in AMPAR trafficking during LTD, even in a subunit-independent manner. Phosphomimetic mutations in GluA1 can affect the endosomal transport of AMPARs during LTD. The association of AP-2 and AP-3 adaptor protein complexes with STG in AMPAR trafficking is important, with the phosphorylation mutation in GluA1 hindering the binding of AP-3 with STG.

C1ql1-Bai3 signaling is necessary for climbing fiber synapse formation in mature Purkinje cells in coordination with neuronal activity

Summary: Changes in neural activity induced by learning and novel environments can lead to the formation of new synapses in the adult brain. This study discovered that Purkinje cells in the cerebellum can be reinnervated by multiple climbing fibers through increased expression of the synaptic organizer C1ql1 or the receptor Bai3. The results indicate that C1ql1-Bai3 signaling, regulated by neuronal activity, may mediate climbing fiber synaptogenesis in mature Purkinje cells.
