Anopheles gambiae odorant binding protein crystal complex with the synthetic repellent DEET: implications for structure-based design of novel mosquito repellents

Insect odorant binding proteins (OBPs) are the first components of the olfactory system to encounter and bind attractant and repellent odors emanating from various sources for presentation to olfactory receptors, which trigger relevant signal transduction cascades culminating in specific physiological and behavioral responses. For disease vectors, particularly hematophagous mosquitoes, repellents represent important defenses against parasitic diseases because they effect a reduction in the rate of contact between the vectors and humans. OBPs are targets for structure-based rational approaches for the discovery of new repellent or other olfaction inhibitory compounds with desirable features. Thus, a study was conducted to characterize the high resolution crystal structure of an OBP of Anopheles gambiae, the African malaria mosquito vector, in complex with N,N-diethyl-m-toluamide (DEET), one of the most effective repellents that has been in worldwide use for six decades. We found that DEET binds at the edge of a long hydrophobic tunnel by exploiting numerous non-polar interactions and one hydrogen bond, which is perceived to be critical for DEET's recognition. Based on the experimentally determined affinity of AgamOBP1 for DEET (K (d) of 31.3 mu Ie) and our structural data, we modeled the interactions for this protein with 29 promising leads reported in the literature to have significant repellent activities, and carried out fluorescence binding studies with four highly ranked ligands. Our experimental results confirmed the modeling predictions indicating that structure-based modeling could facilitate the design of novel repellents with enhanced binding affinity and selectivity.

A Combined Computational Methodology for the Discovery of Hit Compounds with Putative Insect Repellency Properties

Summary: This study aims to discover novel hit compounds for insect repellents by targeting the Odorant Binding Protein 1 (OBP1) using a computational methodology. In vitro and in vivo evaluations showed that coniferyl alcohol and 1,2-diphenyl-2-propanol exhibited high binding affinity to OBP1 and effective repellent activity against Aedes albopictus. These compounds have the potential to serve as scaffolds for the development of novel repellents.

Is the Association of the Rare rs35667974 IFIH1 Gene Polymorphism With Autoimmune Diseases a Case of RNA Epigenetics?

Summary: The IFIH1 gene encodes MDA5, a cytoplasmic RNA helicase involved in innate immunity. The missense variant rs35667974 of IFIH1 is protective for some autoimmune diseases but increases the risk for others. This study shows that the rs35667974 variant affects the catalytic activity of MDA5 in the presence of 2'-O methylation, leading to reduced negative feedback on cytokine signaling and selectively protecting against autoimmunity.

Structural and Biochemical Characterization of the Human Angiogenin-Proliferating Cell Nuclear Antigen Interaction

Summary: The interaction between human angiogenin (hAng) and proliferating cell nuclear antigen (PCNA) was studied using isothermal titration calorimetry (ITC), mutagenesis, and NMR spectroscopy. The two proteins were found to directly interact with each other through in vitro immunoprecipitation studies, and their interaction was quantified using ITC. NMR spectroscopy and molecular dynamics simulations were used to map the interaction surface and construct a structural model for the complex. The validity of the model was confirmed by mutagenesis experiments. Overall, this study provides valuable insights into the structural details and biological roles of hAng and PCNA.

The structure of AgamOBP5 in complex with the natural insect repellents Carvacrol and Thymol: Crystallographic, fluorescence and thermodynamic binding studies

Summary: Among proteins involved in insect olfactory perception, Odorant Binding Proteins (OBPs) are potential targets for discovering compounds to disrupt host-detection behavior. The crystal structures of AgamOBP5 in complex with natural repellents Carvacrol and Thymol, and MPD molecule are presented. Structural analysis revealed that both repellents occupy a binding site with two alternative conformations, and Carvacrol was found bound to a secondary site as well. Fluorescence competition and Calorimetry experiments confirmed the presence of two binding sites and the stabilization effect on AgamOBP5.

Influence of pH on indole-dependent heterodimeric interactions between Anopheles gambiae odorant-binding proteins OBP1 and OBP4

Summary: Insect Odorant Binding Proteins (OBPs) are essential for odor recognition. They undergo conformational changes upon pH change and can form heterodimers with novel binding characteristics. The crystal structures of Anopheles gambiae OBP4 at different pH levels were determined to investigate the pH-dependent heterodimerization mechanism and the interaction with the attractant indole. The results showed that acidic pH induced conformational changes and impaired the binding of indole to OBP4. Molecular Dynamic and Differential Scanning Calorimetry studies further revealed the significant influence of pH on OBP4 stability compared to the effect of indole. Models of OBP1-OBP4 heterodimers at different pH levels were generated and analyzed, suggesting a potential pH-induced heterodimer formation/disruption mechanism.
