The nuclear factor kappa B (NF kappa B) transcription factor plays critical roles in inflammation and immunity. The dysregulation of NF kappa B is associated with inflammatory and autoimmune diseases and cancer. NF kappa B activation is negatively regulated by the ubiquitin-dependent proteasomal degradation pathway. In the present review, we discuss recent advances in our understanding of how ubiquitin ligases regulate the NF kappa B degradation pathway.

Nephrotoxicity Profile of Cadmium Revealed by Proteomics in Mouse Kidney

Summary: This study investigated the proteomic profile changes induced by cadmium in the kidney using a mouse model. It was found that cadmium regulates kidney metabolism, promotes renal damage and cell migration. The downregulation of Na(+)/H(+) exchange regulatory cofactor (NHERF3) could contribute to renal damage, along with a decrease in transferrin (TRF) in the kidney.

Role of autophagy on cancer immune escape

Summary: Autophagy is a catabolic process that degrades intracellular components in lysosomes, potentially affecting cancer cell immune evasion. It may induce degradation of immune checkpoint proteins and manipulate antigen presentation and T cell activity in immune cells. These contradictory effects may stem from different experimental contexts or models.

Role of ferroptosis on tumor progression and immunotherapy

Summary: Ferroptosis, triggered by intracellular iron, leads to cell death through the accumulation of lipid peroxidation. Cancer cells can evade ferroptosis by activating antioxidant signaling pathways. Besides transcriptional regulation, ferroptosis can also be modulated by ubiquitination or autophagic degradation. Induction of ferroptosis enhances immune cell activity, but it may impair T cell activity as well. Therefore, rational combined therapy is crucial for cancer treatment.

Let-7b-5p inhibits colon cancer progression by prohibiting APC ubiquitination degradation and the Wnt pathway by targeting NKD1

Summary: This research aims to investigate the regulatory network of NKD1 and miRNAs in colon cancer. The study identified let-7b-5p as a miRNA that can inhibit colon cancer cell proliferation, migration, and invasion by targeting NKD1. Further analysis revealed that NKD1 enhances colon cancer cell proliferation and migration by inhibiting APC expression.

Inhibition of CK2/ING4 Pathway Facilitates Non-Small Cell Lung Cancer Immunotherapy

Summary: Immune cells play a role in protecting against tumor progression, while aberrant expression of PD-L1 in cancer cells leads to tumor immune escape. This study found that ING4 induces the autophagic degradation of PD-L1, inhibiting immune escape in NSCLC. However, high expression of CK2 was found to correlate with low ING4 protein levels in NSCLC. Further analysis revealed that CK2 induces the phosphorylation of ING4-S150, leading to its degradation by JFK ubiquitin ligase. In contrast, CK2 gene knockout increased ING4 protein stability and T cell activity, inhibiting NSCLC immune escape. Additionally, the combination of a CK2 inhibitor and PD-1 antibody effectively enhanced antitumor immunotherapy.

CD47/SIRPα pathway mediates cancer immune escape and immunotherapy
