Summary: Semaphorins are a group of highly conserved proteins that play a role in axon guidance during neurogenesis and have physiological functions in various organ systems. SEMA7A, an immune semaphorin, regulates diverse immunoinflammatory processes and tumor progression by interacting with specific receptors PLXNC1 and integrins. This review focuses on the role of SEMA7A in autoimmune disorders, inflammatory diseases, and tumors, as well as potential strategies for targeting SEMA7A as a predictive, diagnostic, and therapeutic agent for these conditions.

Summary: This study constructed a large-scale repertoire catalogue of five B cell subsets of patients with immune-mediated diseases (IMDs) and investigated their biological mechanisms. The results showed that autoimmune diseases were associated with a shortened CDR-H3 length in naive B cells and a negative correlation between interferon signature strength and CDR-H3 length, suggesting the role of interferon in premature B cell development. The study also found gene usage skewing in plasmablasts and unswitched-memory B cells of patients with systemic lupus erythematosus (SLE), and this skewing was ameliorated after belimumab treatment.

Summary: This study identified three distinct types of immune checkpoint inhibitor-associated myositis (ICI-myositis) based on gene expression patterns, including ICI-DM, ICI-MYO1 and ICI-MYO2. ICI-DM patients overexpressed type 1 interferon-inducible genes, similar to dermatomyositis (DM) patients. ICI-MYO1 patients had highly inflammatory muscle biopsies and developed coexisting myocarditis.

Summary: CCR2 regulates BCR signaling through the MST1/mTORC1/STAT1 axis. The deficiency of CCR2 reduces the proportion and number of follicular B cells, enhances BCR proximal signaling, and promotes oxidative phosphorylation of B cells. CCR2 deficiency also induces actin filament aggregation and early-activation events of B cells. Glomerular IgG deposition is observed in CCR2-deficient mice, and the expression of CCR2 is decreased in peripheral B cells of SLE patients.
