Summary: This study develops a highly effective acoustic stimulator working at -20 MHz to investigate the roles of acousto-mechanical stimuli in the differentiation of hESCs. Our results show that acoustic dose alters the expression of extracellular matrix and physiological activity-related genes, indicating that acoustic stimulation is an important tool in regulating the stem cells' differentiation processes in cell therapy.

Summary: The study developed a stepwise strategy to generate chemically induced neural crest cells (ciNCCs) and chemically induced corneal endothelial cells (ciCECs) from mouse fibroblasts using defined small molecules. The resulting ciCECs exhibited consistent gene expression profiles and self-renewal capacity to primary CECs, and could be cultured for as long as 30 passages while retaining their features. Transplantation of these ciCECs into an animal model reversed corneal opacity, providing an alternative cell source for regeneration of corneal endothelia and tissues derived from neural crest.

Summary: Periodontal ligament stem cells (PDLSCs) have great potential for neural differentiation, capable of differentiating into various neural-like cells without the need for cell division. They can be induced towards neural lineage using a combination of nutritional factors and have significant implications for neural tissue repair when coupled with biomaterials. PDLSCs can be a valuable target for research and treatment of neurological disorders, such as Alzheimer's disease, multiple sclerosis, and cerebral ischemia. They also show beneficial effects on retinal ganglion cell regeneration and photoreceptor survival, and can be used for peripheral nerve repair through the expression of neural growth factors and differentiation into Schwann cells.

Summary: RNA modifications and their associated proteins play a regulatory role in osteogenesis, and this study provides comprehensive quantitative assessment of these proteins during osteogenic differentiation. The down-regulation of certain RNA modification proteins, mainly involved in methylation and pseudouridylation, was found. Protein-protein interaction analysis revealed their significant associations with osteogenesis-related proteins, and gene set enrichment analysis suggested a potential role of METTL1 in osteogenesis through the cytokine network.

Summary: Stem cell therapy using human umbilical cord Wharton's jelly-derived mesenchymal stem cells (hWJ-MSCs) induced into neurospheres can effectively recover spinal cord injury (SCI) and improve motor function in rats. Neurospheres induced by 10 μM Isx9 for 7 days showed the expression of neural stem/progenitor cell markers through the Wnt3A signaling pathway. Transplantation of these neurospheres into SCI rats resulted in normal movement and tissue recovery, with smaller cavity size in the injury area.
