Control of adipogenic commitment by a STAT3-VSTM2A axis

Summary: This study found that the expression of Vstm2a in preadipocytes is positively regulated by the PI3K/mTOR and cAMP-dependent signaling pathways, but suppressed by the MAPK pathway and the glucocorticoid receptor. STAT3 was identified as a key transcription factor controlling Vstm2a expression in preadipocytes. In mice, elevated STAT3 phosphorylation in the early phases of WAT development strongly correlates with Vstm2a expression.

In prostate cancer patients, TAMs expressing the immunosuppressive marker CD163 independently predict metastases or prostate cancer death. The presence of prostate cancer cells and the cytokine milieu alter the phenotype of TAMs, which may promote tumor growth.

Cis-regulatory hubs: a new 3D model of complex disease genetics with an application to schizophrenia

Summary: The 3D conformation of chromatin plays a vital role in gene regulation, especially in complex diseases. This study discovered functional interactions within cis-regulatory hubs (CRHs) in neurons, providing insights into the epigenetic mechanisms involved in diseases such as schizophrenia. CRHs highlighted significant enrichments in schizophrenia-associated genes, SNPs, and heritability, and also exhibited a higher proportion of differentially expressed genes in schizophrenia compared to other regulatory structures. Therefore, CRHs serve as 3D models to study dysregulation of gene expression in complex diseases.

Small Non-Coding RNAs in Soft-Tissue Sarcomas: State of the Art and Future Directions

Summary: Soft-tissue sarcomas (STS), a rare and diverse group of tumors, have various subtypes characterized by differences in tumor biology and clinical outcomes. Small non-coding RNAs (sncRNAs), including miRNAs and tRNA-derived ncRNAs, play important roles in STS tumorigenesis and differentiation. However, the translation of these findings into clinical applications, such as biomarkers or therapeutic targets, remains challenging. This review summarizes the current understanding of sncRNAs in STS, with a focus on liposarcoma, and discusses their potential as novel biomarkers and therapeutic targets.
