Summary: Untreated HIV-1 infection leads to a decrease in CD4(+) T cell lymphocytes, susceptibility to opportunistic infections, and ultimately death. This study found that cytotoxic lymphocytes can target the HIV-1 Gag protein through granzyme cleavage to potentially control HIV-1 infection.

Summary: The study found that memory-like natural killer (NK) cells and NKG2C-expressing CD8(+) T cells increase in renal transplant patients during cytomegalovirus (CMV) infection, playing a crucial role in combating the CMV virus. The dynamic changes of these cells during CMV viremia may be associated with controlling the virus and preventing graft rejection.

Summary: This study found that CD19(+) B cells in the peripheral blood of multiple sclerosis (MS) patients may express serine-protease granzyme-B (GzmB), resembling cytotoxic CD8(+) T lymphocytes. This finding may provide important clues for investigating the pathogenesis of MS and for the development of new therapeutic interventions.

Summary: The study revealed the critical role of a long noncoding RNA LCDR in regulating lysosome function, which affects lung cancer cell survival. LCDR interacts with hnRNP K to maintain lysosomal membrane integrity, leading to lysosomal cell death and apoptosis. Targeting LCDR/hnRNP K/LAPTM5 axis may provide potential therapeutic targets for cancer treatment.

Summary: X-linked neutropenia (XLN) is caused by gain-of-function mutations in the actin regulator Wiskott-Aldrich Syndrome protein (WASp), leading to reduced numbers of cytotoxic cells in patients. However, NK and T cells from XLN patients show increased granzyme B content and enhanced degranulation and cytokine production compared to healthy controls, indicating an increased capacity to respond to and kill tumor cells.

Summary: The roles of tumor-infiltrating CD4(+)Foxp3(-) T cells have been further clarified in a mouse model of colon cancer. These cells maintain an effector phenotype, express inflammatory cytokines and cytolytic molecules, and exhibit cytotoxic function against tumor cells. They are distinct from CD4(+) regulatory T cells and are activated through the IL-12/IL-27 pathway. This study highlights the importance of understanding the plasticity and functional diversity within the CD4(+) T cell population in the tumor microenvironment.

Summary: This study evaluated the early immune response of recipients with disparate HCMV outcomes and found that HCMV-specific IFN-gamma secreting cells, HCMV IgG, and absolute lymphocyte count could represent the humoral and cellular immune response. Early monitoring of these immune markers could enable prediction of HCMV outcomes posttransplant and assessment of the severity of HCMV DNAemia.

Summary: This study characterized age-associated alterations in immune cells across multiple mouse organs and identified a distinct population of age-associated GZMK-expressing CD8(+) T cells. These Taa cells, highly clonal with specific features, were found to be a potential target to address age-associated dysfunctions of the immune system. In humans, proportions of the circulating GZMK(+) CDE3(+) T cell population that shares signatures with mouse Taa cells also increased during healthy aging.

Summary: In this study, the researchers aimed to investigate the presence and function of T-RM cells in healthy and allograft kidney tissue by analyzing the phenotype and function of T cells through multichannel flow cytometry. The results showed that kidney tissue samples contained a significant number of CD8 and CD4 T cells that were more actively cycling, expressed specific markers CXCR3 and CXCR6, produced higher levels of IFN gamma, and displayed polyfunctionality compared to circulating T cells.
